M1균주에서의 10μM에서 30 μM농도의 Congo red, Acid Yellow 9 및 Orange II 투입시는 일주일 배양후 거의 95%이상 분해가 되었으나, WT균주는 50%에서 80%까지만 제거되었다. 균주 종류 및 투입시기에 상관없이 Congo red, Acid Yellow 9, Orange II순서로 분해속도가 빨랐다.

This study was aimed to develope a method for degrading azo dyes to that 50% of dye-containing wastewater are attributed by using a white-rot wood decaying basidiomycete, Phanerochaete chrysosporium. Recent researches have been mainly focused on elucidation of reaction mechanisms of lignin peroxidase(LIP) and Manganese peroxidase(MnP) so that little application efforts have been made. A major problem in using LiP and/or MnP for degrading aromatic dyes was that activities of these two enzymes could not be maintained for a longer operation. Therefore, culture conditions of a wild type(WT) and a mutant of P. chrysosporium for maximal LiP activity were established. In addition, Mn^2+, concentration in the culture medium and a timing of dye input were optimized. Higher levels of lignin peroxidase activitity were obtained with 0.2M 2,2-dimethyl succinate buffer solutions and 150 rpm. Concentrations of 0.4mM veratryl alcohol, 12ppm MnSO₄·H₂O and 0.05% Tween 80 gave optimal lignin peroxidase activities in this medium. Maximum lignin peroxidase activity was 53.8 U/L for P. chrysosporium wild type and 792.4 U/L for mutant M1. In this experiments, dye addition time was 4 day(wild type) and 8 day(mutant M1) for a maximum dye decolorization rate. The P. chrysosporium mutant M1 decolorized 10 μM and 30 μM dyes more than 95% and the wild type decolorized more than 50 %. Dye decolorization rate was in the order of Congo red, Acid yellow 9 and Orange II.
