When a foreign surface comes in contact with blood, the initial blood response is adsorption of blood proteins, followed by platelet adhesion and activation of the coagulation pathways, leading to thrombus formation. A particularly effective polymer for the prevention of protein adsorption and platelet adhesion appears to be polyethylene oxide (PEO), probably due to its minimum interfacial free energy with water, hydrophilicity, highly surface mobility and steric stabilization effects, and unique solution properties and molecular conformation in water. In this study, water-insoluble terpolymers with pendant PEO and negative charge groups as blood-compatible coating materials were synthesized by MMA, MPEOMA (PEO mol. wt., 1,000), and vinyl sulfonic acid sodium salt (VSA). MMA is hydrophobic and provides the terpolymers to be water-insoluble by controlling monomer ratio. MPEOMA is hydrophilic and provides the terpolymers blood-compatible due to the PEO chain's high mobility and steric stabilization effect in water. Hydrophilic and negatively chargeable VSA can also provide blood-compatible by the electrostatic repulsions of blood components. PEO- and negative charge-containing surfaces were prepared by spin coating of the synthesized MMA/MPEOMA/VSA terpolymers in PU or PMMA matrix. The surface properties were investigated by the measurements of water contact angle and Zeta-potential. The water contact angles of the terpolymer surfaces decreased with increasing MPEOMA or VSA content in the terpolymers, due to the hydrophilic property of those exposed onto the surfaces. The behavior of plasma protein adsorption and platelet adhesion on the terpolymer-coated surfaces was compared to that on the surfaces coated by MMA/MPEOMA or MMA/VSA copolymers. The MMA/MPEOMA/VSA terpolymers (monomer molar ratio, 8/1/1 or 7/1.5/1.5) were particularly effective in preventing plasma protein adsorption and platelet adhesion on the surfaces, probably due to the synergistic effects of PEO and negatively charged groups. And PU tubes as vascular grafts (diameter 5mm) were prepared using a spinning method and the insides of tube s were coated with the synthesized terpolymers using a same method. Finally, PU tubes coated with the terpolymers which have effective blood-compatibility will be used ex-vivo experiment. Hence, terpolymer coated PU tubes will be evaluated application ability as vascular graft.
