LPC 투여 시 Ca^(2+) 전류의 막전압 의존성 활성화 곡선이 오른쪽으로 일부 이동하였으나 막전압 의존성 비활성화 곡선에는 변화가 없었다. 4. LPC에 의한 Ca^(2+) 전류 증가 효과는 protein kinase A (PKA) 촉진제인 forskolin 전처치에 의해서는 영향을 받지 않았으나 protein kinase C (PKC) 촉진제인 phorbol myristic acid (PMA)를 전처치하는 경우 소실되었다. 5. PKC inhibitor인 staurosporine이나 chelerythrine을 전처치하는 경우 LPC에 의한 Ca^(2+) 전류 증가 효과가 소실되었다. 이상의 실험 결과로 볼 때 LPC에 의한 Ca^(2+) 전류의 증가 효과는 세포 내 PKC 활성변화를 매개로 하여 이루어지며, 이에 의한 평활근 세포 내 Ca^(2+) 농도 증가 효과는 죽상경화증 혈관에서 관찰되는 혈관운동 기능이상의 발생에 중요한 역할을 한다고 생각된다.

Impairment of relaxing response and augmentation of contractile response to vasoactive substances have been reported in atherosclerotic arteries. These alterations in vascular reactivity are considered as an underlying mechanism for the development of acute vasospasm in atherosclerotic artery. Recently, it has been reported that lysophophatidylcholine (LPC), an oxidative metabolite of low density lipoprotein causes this functional abnormality. However, the precise mechanism of LPC induced change of vascular reactivity is still uncertain. In this study, to elucidate the underlying mechanisms of abnormal vascular reactivity in atherosclerotic artery, we directly examined the effect of LPC on whole cell Ca^(2+) current using patch clamping technique. Application of LPC (1μM) induced a significant increase in I_(Ca) (142±9%, p<0.01), which was readily reversed by washout of LPC. Steady state voltage dependency of activation or inactivation properties of I_(Ca) was not significantly changed by LPC. The PKC inhibitor, staurosporine (10 nM) or chelerythrine (3μM) inhibited I_(Ca) by 54±6% (p<0.01), 47±6% (p<0.01), respectively. After inhibition of I_(Ca) by PKC inhibitors, application of LPC had no significant change in the amplitude of I_(Ca). The PKC activator, phorbol myristic acid (PMA, 100 nM) stimulated I_(Ca) by 124±3% (p<0.01). After stimulation of I_(Ca) by PMA, application of LPC had no significant change in the amplitude of I_(Ca). These findings suggest that LPC increase I_(Ca) by a pathway that involves endogenous PKC in vascular smooth muscle cells, and LPC-induced increase of I_(Ca) might be, at least in part, responsible for development of abnormal vascular reactivity in atherosclerosis.
