밀, 쌀, 옥수수와 감자로부터 분리, 제조한 생전분과 가열-냉각 과정으로 처리한 RS3, 화학적 가교결합으로 제조한 RS4 저항전분(RS, Resistant starch)으로 RS 함량을 분석하고 특성을 비교하였다. In this study, RS levels of raw, autoclaving-cooling cycled RS3, and chemically modified RS4 from wheat, rice, corn and potato starches were analyzed and their morphological characteristics were compared. P/G(pancreatin-gravimetric), AOAC, and Englyst et al's methods were used for RS analysis and morphological properties and crystallinities of residues after hydrolysis of RS were investigated by Scanning electron microscopy and X-ray diffractometery. RS levels of mixtures of two of three type starches, raw, RS3 and RS4 starches, with same weight ratio were analysed and compared with prediced values. RS levels of wheat flour added 20%(w/w) different type resistant starches were also measured using P/G and AOAC method. RS levels of raw and RS3 starches measured were slightly higher in P/G method than in AOAC method regardless of starch source. In the case of RS4, RS levels by AOAC method were much higher than those by P/G method. Experimental and predicted RS levels measured using P/G method were almost similar, whereas experimented and predicted RS levels by AOAC method were slightly different. RS levels of wheat flours with different type resistant starches(20% with flour basis) showed same tendency with those of each type starch. After resistant starches were digested with enzyme, the residues were shown different shape with assay methods. All kinds of residues after hydrolysis were completely degraded and ruptured using P/G method, but using UP/G method, starch granules were partially degraded with maintaining their original shape. Using AOAC method, raw and RS3 starches were digested but RS4 starches kept the original shape having some pores. By Englyst method, RS3 were digested but raw starch granule and RS4 starch maintained original shape. On X-diffractogram, raw starch of wheat, rice and corn starches showed typical A type which was shown peaks at 2θ=15, 17, 23.5˚. RS residues using UP/G and Englyst methods also showed same peak as raw starch. While RS starches were digested using P/G and AOAC methods, the peaks were disappeared. Raw potato starch showed typical B type which was shown peaks at 2θ=5, 17.2˚. RS3 of all starches showed B type after digesting.
