To identify the main antioxidative components of black soybean, the contents of isoflavones, phenolic acids, and tocopherol were measured and compared their antioxidative activities with yellow soybeans. Black soybean has been widely utilized as oriental medicine as well as food because of its unique functionality. Antioxidative activity of 80% methanol extract of black soybean(Cheongjakong) was measured by TEAC assay and compared with yellow soybean(Danwonkong). As a result, black soybean showed 28% higher activity than yellow soybean. Even though the contents of isoflavones, phenolic acids and tocopherol in black soybean measured by reverse-phase HPLC were lower than yellow soybean’s except anthocyanin, black soybean showed stronger acitivity than yellow soybean. Anthocyanin, which is a main pigment in the seed coat of black soybean, might be responsible for its antioxidative activity. The compounds of anthocyanins contained in the test black soybean(Cheongjakong) were identified as cyanidin-3-glucoside, delphinidin-3-glucoside, and petunidin-3-glucoside and the contents were 1.42, 5.77, and 0.3mg/g respectively. To compare antioxidative activities of the main components containing in the black soybean, antioxidative effects were measured by TEAC assay and DPPH assay using the commercial standards of anthocyanin, isoflavones, phenolic acids and tocopherol at the same level existed in black soybean. The most effective component was gentisic acid and followed by anthocyanin, p-coumaric acid, ferulic acid, genstein, syringic acid, daidzein on TEAC assay. On the free radical scavenging activity measured by DPPH assay, gentisic acid showed the strongest effect and followed by anthocyanin, syringic acid, ferulic acid, chlorogenic acid, but genistein showed very low effect. In addition, the defense abilities of these components against oxidation on the homogenates of liver and skin of mice fed vitamin E-deficient diet were evaluated TBARS and protein carbonyl value after provocation by UVB radiation. As a result, genistein, gentisic acid, γ-tocopherol, and anthocyanin showed strong protective effects on mice tissue oxidation, though its activities were different according to UVB exposure duration. Although these components had synergistic effects each other, anthocyanin showed the strongest effect especially. To identify the main antioxidative component of black soybean, 80% methanol extract of black soybean was fractionated by hexane, chloroform, ethyl acetate, butanol and water according to their polarity. Among these solvent extractions, the ethyl acetate fraction showed the strongest antioxidative activity and the highest total phenol content. Although anthocyanin usually contained in the chloroform fraction, it was spread other fractions too. The strongest antioxidative effect of ethyl acetate fraction might be responsible for its phenolics and anthocyanin, which be cooperated to show synergistic effect and distribute to suppress lipid peroxidation.
