NO synthesis is beneficial or protective to the liver during systemic sepsis. However, the mechanism by which NO can play an anti-inflammatory role in vivo is unclear. This study on NO in the liver stems from an interest in the mechanism leading to the altered liver function in sepsis or inflammation. Thus, the effect of endogenous NO on the mice liver damages and the effects of serum and ionizing irradiation on NO production in embryonic mouse liver cell line BNL CL.2 were studied to establish the role of NO in the liver under sepsis or inflammation. Roles of NO and tumor necrosis factor - α(TNF-α) in liver inflammation induced by Corynebacterium parvum(C. parvum) and LPS was investigated. C. parvum-treated mice produced large amounts of NO and developed a severe liver injury in response to LPS. Concurrent adminstration of aminoguanidine(AG), non-selective NO inhibitor, following LPS exposure was not only suppressed NO synthesis in these animals but also increased the hepatic damage. To elucidate NO production hepatoprotective mechanism, effects of superoxide dismutase (SOD) and TNF-α content of serum were investigated. Treating the mice with SOD could reduce the hepatic damage induced by inhibition of NO production during endotoxemia. TNF-α content of serum peaked at about 3 hr after LPS injection. In contrast, NO content of serum peaked by 5 hr after LPS injection. The treatment of the mice with AG led to enhanced TNF-α production. These results indicate that NO serves a protective role in the liver and reduces hepatic injury in endotoxemia. This protective action may be mediated by the capacity of NO to neutralize superoxide and to Suppress LPS-induced TNF-α production. The fact that when BNL CL.2 cells, were cultured with serum-free medium, they were induced to produce NO by the stimulation of interteron-γ (lFN-γ) alone or IFN-γ plus LPS was demonstrated. Serum-starved cells showed significant amount of nitrite production and iNOS protein expression in response to IFN-γ in dose- and time- dependent manners, but serumsupplied cells did not. The production of NO was blocked by protein tyrosine kinase(PTK) inhibitors, genistein and herbimycin, but not by protein kinase A or C inhibitors. These results suggest that the deprivation of serum from BNL CL.2 cell culture medium might prime the cells to produce NO when the cells are triggered by IFN-γ and the involvement of PTK signal transduction pathway in the expression of iNOS gene in mouse hepatocytes. The production of NO in BNL CL.2 cells, following exposure to ionizing radiation in vitro, was investigated. High-dose(6 Gy) irradiation induced NO production from BNL CL.2 cells treated with lEN-γ alone or IFN-γ plus LPS. Because ROI is produced in response to ionizing radiation, the participation of ROI in NO production was examined. After ionizing radiation, NO production was not affected by treatment with SOD, but apparently decreased by the treatment with catalase(CAT), an enzyme which can convert H_2O_2 to water, or SOD plus CAT. These results suggest that radiation-induced production of NO in BNL CL.2 cells might be attributed to H_2O_2 formed by ionizing irradiation.
