Methanol extracts from 63 plant species were tested for their acaricidal activity against Dermatophagoides farinae (Hughes) and Dermatophagoides pteronyssinus (Trouessart) using direct contact method. The larvicidal activity against Aedes aegypti (L.) and Culex pipiens pallens (Coquillett) were examined using dry film method. Responses varied according to plants, mites, and mosquito species. After 24 hr exposure, potent acaricidal activity against two mites was produced from the extract of Chamacyparis obtusa leaves (family Cupressaceae) at 10 μg/㎠, and the extract of C. obtusa revealed potent larvicidal activity against two mosquitoes at 50 ppm. Because of potent acaricidal and larvicidal activity of methanol extract of C. obtusa against two mites and mosquito larvae, the extract of C. obtusa was sequentially partitioned with hexane, chloroform, and water portions. The hexane portion exhibited potent activity, but chloroform and water portions did not show any activity. Purification of the biologically active constituents from hexane portion was done by using silica gel chromatography and high performance liquid chromatography. The structure of acaricidal and larvicidal compound was by ¹H-NMR, ^13C-NMR and Mass spectrum, and identified as hinokitiol (β -thujaplicin C_10H_12O₂). The acaricidal and larvicidal activity of hinokitiol was compared with that of the widely used benzyl benzoate and N,N-Diethyl m-toluamide (DEET). Based on 24 hr LD-50 value, acaricidal acitivity against D. farinae was more pronounced in hinokitiol (7.25 μg/㎠) than benzyl benzoate (8.54 μg/㎠) and DEET (37.59 μg/㎠). Acaricidal acitivity against D. pteronyssinus was more pronounced in hinokitiol (6.22 μg/㎠) than benzyl benzoate (6.68 μg/㎠) and DEET (17.98 μg/㎠). Larvicidal acitivity against A aegypti was more pronounced in hinokitiol (25.22 ppm) than benzyl benzoate (26.54 ppm). Against C. pipiens pallens larvae, larvicidal acitivity was more pronounced in hinokitiol (22.52 ppm) than benzyl benzoate (23.68 ppm). Typical poisoning symptoms of hinokitiol to D. farinae, D. pteronyssinus, A aegypti, and C. pipiens pallens were compared with those of benzyl benzoate and DEET. All compounds resulted in lethargy of treated mites and mosquito larvae, being directly connected to death. Based on these data, the naturally occurring C. obtusa leaf-derived compound could be of practical use as D. farinae, D. pteronyssinus, A. aegypti, and C. pipiens pallens control fumigants for a relatively long time, provided that a carrier producing a slow-release effect can be selected or developed.
