본 연구에서는 알킬(alkyl) 사슬들을 도입한 페네틸아민(phenethylamine) 유도체들의 합성, 항균 활성 측정을 통해 구조-활성 상관관계가 수행되었다. 짧은 길이의 탄소수 2∼5의 알킬기와 긴 길이의 탄소수 16∼18의 알킬기가 치환된 페네틸아민 유도체들은 활성이 없었고 반면에 탄소수 7∼14의 알킬기가 치환된 페네틸아민 유도체들은 활성이 향상되었다. 페네틸아민 유도체들은 그람 양성세균(Gram-positive bacteria), 그람 음성세균 (Gram-negative bacteria), 그리고 곰팡이(fungi)의 일종인 칸디다(Candida)에 대해 광범위한 활성을 나타내었다. 그람 양성세균과 곰팡이에 대한 활성은 그람 음성세균보다 더 좋은 활성을 나타내었다. 특히 페네틸아민 유도체중에서 탄소수 12의 도데실(dodecyl)기를 가진 도데실 페네틸아민(N-dodecylphenethylamine), 도데실 타일아민(N-dodecyltyramine), 그리고 4-메톡시도데실 페네틸아민 (4-methoxy-N-dodecylphenethylamine)은 S. epidermidis, B. subtilis, 그리고 E. coli 에 대하여 카나마이신(kanamycin)에 비해 1/2-1/4배 활성을 나타내었으나 E. faecalis 에 대해서는 2-4배 높은 활성을 나타내었다. 또한 도데실 페네틸아민이 카나마이신과 비교하여 그람 음성세균인 K. pneumoniae(MIC 4)에 대해서는 좋은 활성을 갖는 것은 주목할 만하다. 실험한 유도체들 중에서 P. mirabilis 와 P. aeruginosa 의 세균들에 대해서는 아무런 활성도 없었다. 이러한 결과는 어떤 구조의 소수성(hydrophobic) 치환체들의 존재와 크기가 최적의 항균활성에 결정적이라는 사실은 제안한다. 페네틸아민 방향족고리(aromatic ring) 에 산소(oxygen) 치환효과를 보기위해 하이드록실(hydroxyl)기를 도입한 경우, 항균활성이 할로젠(halogen)원소들이나 메톡시(methoxy)기, 메틸(methyl)기를 도입한 유도체들보다 뛰어난 활성을 나타냈다. 합성된 화합물들의 항진균에 대한 활성은 메톡시(methoxy)기를 갖는 페네틸아민 유도체들이 여러치환체를 도입한 페네틸아민 유도체들 보다 좋은 활성을 나타내었다. 한편, 데실기, 도데실기를 도입하여 합성된 벤질아민(benzylamine)과 싸이클로헥실에틸아민(cyclohexylethylamine)유도체들은 페네틸아민 유도체들에 비해 낮은 항균활성 나타내었다. 페네틸아민의 특성연구에 있어서 리포솜(liposome)을 이용한 실험은 도데실기를 갖는 페네틸아민 유도체가 세포막과 상호작용에 의한 막의 교란을 통해 미생물을 저해할 수 있음을 제안할 수 있다. The syntheses of phenethylamine derivatives with alkyl chains and their in vitro structure-biological activity relationships are discussed. Introduction of shorter alkyl (C1 to C5) chains and longer alkyl (C16 and C18) chains to the phenethylamine resulted in no antimicrobial activity. While phenethylamine derivatives having heptyl, octyl, nonyl, decyl, undecyl, dodecyl, and tetradecyl (C7 to C14) chains significantly enhanced the activities. Activity against the Gram-positive bacteria and fungi proved to be much higher than against the Gram-negative bacteria. In particular, N-dodecylphenethylamine, N-dodecyltyramine, and 4-methoxy-N-dodecylphenethylamine showed less inhibitory activity against S. epidermidis, B. subtilis, and E. coli by 1/2-1/4 than kanamycin sulfate, while they were 2-4 times more active than kanamycin sulfate against E. faecalis. It is noteworthy that N-dodecylphenethylamine possessed stronger activiy against Gram-negative bacterium K. pneumoniae (MIC 4) than kanamycin sulfate. None of the derivatives showed activtiy against P. mirabilis and P. aeruginosa. This result suggested that the presence of lipophilic substutuents of certain structures and sizes might be crucial for the optimal antimicrobial activity. The effects of the oxygen substituents on aromatic ring of phenethylamine on the antimicrobial activity were also discussed. The position for introduction of hydroxyl group and methoxy group on the aromatic ring made marked difference in the effect on the antimicrobial activity. N-alkylphenethylamines bearing the substituent OH in the para position exceeded the antibacterial activity of the N-alkylphenethylamines with F, Cl, Br, OCH₃, and CH₃ residues the aromatic ring. The antifungal activity of synthesized compounds, revealed that N-alkylphenethylamines bearing the substituent OCH₃ in the para position exhibited the highest activity against Candida species. Benzylamine derivatives and cyclohexylethylamine derivatives having decyl, and dodecyl (C10 and C12) chains showed antimicrobial activities. The leakage of dye from liposomes induced by N-dodecylphenethylamine suggests that antibacterial action of this compound are due to its interaction with and perturbation of the bacterial membrane. Quantitative structure-activity relationship (QSAR), using genetic function approximation (GFA) technique was used to examine the correlations between the calculated molecular descriptors and the in vitro activities of a series of phenethylamine derivatives. Model generated for phenethylamine derivatives against B. subtilis show that thermodynamic factor as expressed by logP and structural factor as expressed by H-bond donor are important whereas for phenethylamine derivatives against C. albicans, logP and topological property accounted for by kappa index order 2 play an important role for the activity.
