The objective of this research is the development of an analytical system for high-density lipoprotein (HDL) cholesterol, as an indicator of the incidence of coronary heart disease (CHD), enabling for minimal-invasive diagnosis. A Conventional system with stacked membrane pads for HDL cholesterol required approximately 30 to 50 μL sample drawn from a peripheral blood by pricking the body skin. To alleviate a burden of pain to human body, we pursued two different way of sample transportation, from the sample application port to the detection site of the final signal, minimizing dilution and loss of the sample. We, first, devised an analytical system employing a membrane strip made of glass fiber as a way of sample migration. The use of this membrane was beneficial for gaining a fast absorption rate of aqueous medium and low adhesion of analyte, i.e., plasma lipoprotein. As an alternative approach, we also constructed a system with micro-fluidic plastic channel with similar properties to that of the membrane but relatively defined structure. For point-of-care testing, HDL cholesterol has been conventionally assayed using vertically stacked membrane pads with each unique function required for performing the cholesterol assay. Once the sample was applied on the top, the medium was migrated to the bottom along the gravity. We developed such along-the-gravity medium flow system for the analysis of HDL cholesterol as conventional, standard format. To achieve a reduction of sample volume, a glass membrane strip was adopted additionally for transferring a small sample to a vertically stacked of membrane pads, similar to the standard but with the reversed stacking arrangement, and then sample was flown against the gravity by absorption. Since this mode of medium flow was relatively slow, enzyme reactions to decompose cholesterol proceeded for an extended time increasing the signal yield. We assessed analytical performances of the against-the-gravity medium flow system by comparing with those of the conventional. Although the against-the-gravity flow system employed five times smaller sample volume for the assay, the performances were not significantly different from that of the conventional towards analysis time, reproducibility of measurements, capability of HDL separation, and dynamic range. To investigate the other version of assay system offering a potential of further reduction of sample volume, a micro-fluidic system was developed utilizing a defined channel containing membrane pad inside as solid matrix for protein immobilization. With such system developed, the sample size was able to be reduced down to 2 μL, maintaining the dynamic range acceptable for clinical assays for HDL cholesterol. A drawback in the assay was the production of a weak signal comparing to that of the conventional due to the dilution of sample and loss in separating HDL from other lipoproteins. This system also showed a low reproducibility in repetitive measurements, which may be caused by a manual sample handling and sub-optimal operating conditions. In this research, we devised analytical systems for HDL cholesterol with enhanced performances particularly in handling small sample sizes utilizing a glass fiber membrane strip and micro-fluidic plastic channel as alternative ways of sample migration. Comparing to the conventional, we were able to reduce the sample volume maximum 8 times without significantly sacrificing the analytical performances.
