Theoretical studies of the sodium cation binding and isotope hydration effects on the model compound B-DNA ($C_pG_pC$) have been fully elucidated by using empirical potential energy functions. In the first place the $Na^+$ cations bound to phosphate anions have been optimized and the average distance Na...O(phosphate anions) has given 2.18 $\mbox{\AA}$ which is in a good agreement with ab initio SCF computations. The hydration scheme of this model compound has been obtained by minimizing the interaction energy between water molecules and the substrate and it has been turned out that the total 17 water molecules are hydrated to B-DNA, i.e., four water molecules to each sodium cation, two to oxygens in phosphate and sugar and the others to nucleosides. Also the isotope hydration due to substitution of $D_2O$ for $H_2O$ has been carried out by the same procedure. Here the major factor to control over the isotope effect on the hydration have been known as the electrostatic contribution. Finally, the net stabilization of B-DNA has been explained and compared with in terms of the stabilization due to the hydration and the cation binding energy, i.e., the net interation energy is -299.3 Kcal/mol compatible with five times as much as that in $Na^+$ free model compound.
