A large number of Alzheimer’s disease (AD) researches are focused on tau phosphorylation and β-amyloid (Aβ) toxicity. Hence, the molecular identity of the enzyme involved in tau phosphorylation of AD has been debated. Here, I showed that inactivation of protein phosphatase 2A (PP2A) rather than activation of glycogen synthase kinase (GSK3β) is involved in tau phosphorylation of degenerating neurons treated with methotrexate or okadaic acid (OA). Emerging evidence suggests that not only Aβ but also other amyloid precursor protein (APP) fragments, such as the β-C-terminal fragment (βCTF), might be involved in AD. Interestingly, APP-βCTF was accumulated in the axonal swellings of OA-treated neurons, which are also filled with various cytoskeleton constituents and motor molecules similar to the early pathology of AD. This APP-βCTF was phosphorylated at Thr668 and colocalized with phosphorylated JNK and JIP-1/3 implicating pathologic formation of APP-JNK-JIP complex. Searching for the intracellular compartment of generating APP-βCTFs, upregulation of lysosomal system including autophagy was found in OA-treated neurons and substantial colocalization of APP-βCTFs with autophagosome was observed. In according to the increase of APP-βCTF, β-secretase (BACE-1) also increased and BACE inhibitor reduced the axonal accumulation of APP-βCTF and showed protective effects in OA-induced neurodegeneration. Surprisingly, similar results were observed with an intracellular zinc chelator, N,N,N′,N′-tetrakis(2-pyridylmethyl)ethylenediamine (TPEN). These findings suggest that intracellular zinc might be dysregulated in OA-treated neurons and that this could contribute to the amyloidogenic processing of APP. These protective effects of BACE inhibitor and TPEN involved the calpain pathway. This study suggests that PP2A is a pivotal enzyme involved in AD pathogenesis and that regulation of APP processing through drugs controlling BACE-1, zinc and calpain could be protective in AD.
