Objective: The objectives of this study were as follows; (1) to analysis gene expression in PAH exposed and smoking groups compared with controls, (2) to examine carcinogenic mechanism in tobacco components treated lung cells, and (3) to evaluate usefulness of microarray-based genomic survey which is a high-throughput approach to parallel study on expression patterns of numerous genes. Materials & Methods: For human clinical study, 2,000 workers who had visited to Korea University Hospital for an annual health examination, were enrolled in our study, and they were divided into two groups as experimental and control groups according to smoking habits and urinary 2-naphthol levels, which were examined using by high performance liquid column (HPLC). For in vitro study, human fetal lung fibroblasts (WI38) and human lung adenocarcinoma (A549) were cultured, and they were treated with tobacco compounds such as nicotine, benzo[a]pyrene, and 2-Naphthylamine. We analyzed gene expression profiles using cDNA microarray and real time RT-PCR in human subjects and lung cell lines. Results: In human study, the expressions of genes related with inflammation and cell proliferation were up-regulated and those of genes related with DNA repair and apoptosis were down-regulated in both smoking groups and PAH exposed groups. In vitro study, the gene expressions related with inflammation, proliferation were also up-regulated, and those related with apoptosis, immune response were down-regulated in both WI38 and A549 cell lines treated with three tobacco compounds. Real time RT-PCR showed significant increases in Ahr, Arnt, AKR1B10, CYP1A1, and CYP1B1 gene expression in tobacco compounds-exposed samples. Conclusion: We observed that tobacco compounds could influence apoptosis, inflammatory, immune, and cell cycle of human subjects and lung cells. From these results, we suggested that these metabolic changes caused by smoking or tobacco compounds should explain the relationship between smoking and lung cancer and be due to increased gene-expression of enzymes or receptors related with tobacco compounds metabolism. The microarray-based genomic survey is a high-throughput approach for evaluation of gene expression, characterization of tobacco compounds.
