Severe acute respiratory syndrome-coronavirus (SARS-CoV) causes an infectious disease through respiratory route. I cloned and monitored the expression of the SARS associated coronavirus (CoV) spike S1-RBD region. The spike (S) protein of SARS-CoV consists of S1 and S2 domains are responsible for virus binding and fusion, respectively. Receptor-binding domain (RBD) positioned in S1 can specifically bind to angiotensin-converting enzyme2 (ACE2) on target cells. I raised monoclonal antibodies (mAb) against the SARS associated coronavirus spike protein S1 RBD domain. The recombinant RBD domain of SARS-CoV spike protein has been successfully elicited SARS-CoV RBD specific IgG antibody in mouse after immunization with purified recombinant RBD protein. The RBD-specific mAbs may be useful for further study of the function of the S protein and for diagnosis of SARS-CoV infection. It is also important for hypertension, because angiotensin-coverting enzyme2 (ACE2) serves as a receptor for SARS. ACE2 might regulates the balance between vasoconstrictors and vasodilators within the heart and kidney. It was proved that the recombinant RBD binds to target cells such as mouse heart, lung, and kidney cells through ACE2, and this result can be applied for the future research to develop a vaccine for SARS and to develop a drug regulating blood pressure.
