GAD 효소 조정제에 의한 최적 생물전환에서 고농도 MSG 전구체 (200 g/L)를 사용한 GABA의 생물전환 생산실험을 통하여 100 g/L 이상의 GABA 생산하였다.

In this study we isolated 690 strains of lactic acid bacteria (LAB) from kimchi, yoghurt, soy pastes and fish pastes. Selected 690 strains were cultured in a MRS medium containing 5% and 10% (w/v) monosodium glutamate (MSG) by incubating at 30℃ for 48 hours. The strains grown in the medium with high concentration of MSG were cultured in the liquid medium containing MSG and measured the productivity of GABA using thin layer chromatography (TLC) and HPLC methods. Selected strain BFC-110 was identified asg thin layer chbrevis on the basis of morphology and 16s rRNA homology, The biotransformation conditions of MSG to GABA were optimized with the cheaper industrial medium instead of MRS. Under the optimized culture conditions, the yield of GABA reached 9.587 g/L (with MSG) and 2.069 g/L (without MSG), in flask scale. The yields of GABA in optimized 5L, 50L, and 300L fermenter scale reached 26.7, 20.0 and 20.3 g/L of respectively. Bioconversion rate were 95, 73 and 92% in 5L, 50L and 300L fermentation when BFC-110 was fermented in the optimal medium containing 2% maltose, 3% yeast extract, 0.2% ammonium citrate, 0.01mM MgSO4, 0.5% sodium acetate, 0.2% dipotassium hydrogen phosphate at 30℃ with initial pH 7.0. Furthermore, after optimizing the condition of enzyme conversion with GAD, bioconversion rate was above 100g-MSG/L.
