The effects of three extraction methods (CTAB, magnetic bead and silica gel membrane methods) on the yields and quality of genomic DNA extracted from corn, soybean and theirs processed foods were assessed. Among the three different extraction methods, the highest DNA extraction yields from corn, soybean and theirs processed foods were obtained using the CTAB method. However, the silica gel membrane method resulted in the better DNA purity than the other methods in corn and its processed foods, determined by the ratio of absorbance at 260 to 230 nm. And the CTAB method resulted in the better DNA purity than the other methods in soybean and its processed foods, determined by the ratio of absorbance at 260 to 280 nm and 260 to 230 nm. According to cycle threshold (Ct) values of RT-PCR, the silica gel membrane and the CTAB method were capable of extracting the highest quality of genomic DNA from corn, soybean and theirs processed foods respectively. Therefore, the silica gel membrane method and the CTAB method were determined to be the most suitable for extracting genomic DNA from corn, soybean and theirs processed foods respectively. This method may be employed to prepare for genomic DNA from corn and soybean products for the efficient detection of extraneous genes in genetically modified (GM) foods.
