Bacillus sp. KN-517에 의한 keratinase의 분자량 및 순도를 확인하기 위하여 SDS-PAGE를 한 결과 단일 band를 보였으며 분자량은 약 28 kDa이었다. 또한 모발을 탄소원으로 사용 시 120시간 만에 효소활성이 88 units/mL/min에 도달하였으며, 이 때 모발은 대조군에 비하여 20% 정도 가늘어졌다. In this work, the application possibility on the hair cosmetics of keratinase was investigated by decomposing whole chicken feather and optimizing keratinase production. The microbial strain producing keratinase in a high activity was selected and identified. The isolated strain was identified as Bacillus sp. KN-517 based on its morphological and biochemical characteristics. The optimal conditions for the production of keratinase by Bacillus sp. KN-517 were the 3.5% chicken feather as a nutrition source, 1.0% tryptione as an organic carbon source, 0.5% KNO3 as an inorganic nitrogen source, and 0.05% KCL, 0.03% K2HPO4, 0.04% KH2PO4 as a mineral source and, 0.01% yeast extract as a growth factor. The optimal temperature and pH were 40°℃ and 8.5 with shaking culture (200 rpm/min), respectively. The molecular weight and purity of the keratinase produced by Bacillus sp. KN-517 showed 28 kDa single band and respectively, from the results of the SDS-PAGE. For the use of hair as a carbon source, enzyme activity was reached to 88 units/mL/min only for 120 hours and its thickness was decreased. Keratinase produced from Bacillus sp. For the treatment of enzyme in a long time and high temperature, partial decomposition of keratin protein of hair and cleaning of hair surface were improved. After the pre-treatment of keratinase produced from Bacillus sp. KN-517 to the thick hair or thin hair respectively, the possibility for the application of hair cosmetics for the generation of effective permanent wave by using thioglycolic acid as the permanent wave reducing agent was studied. For the evaluation of the kratinase effects, the results of the pre-treatment of keratinase to the thick hair and thin hair and successive permanent were compared. Its effectiveness was measured by the change in the thickness, tensile strength, MB dying, wave shape, wave continuance, and SEM. The formation rate of wave for the thick hair and thin hair by enzyme treatment was significantly high compared to those without treatment due to the relatively narrow distance. The formation rate of wave for the thick hair was relatively higher than that of thin hair. The duration of the waves continuance was relatively high for the enzyme treatment of thick hair deposited by thioglycolic acid and subsequently exposed 20 minutes in the natural conditions. Therefore, high formation and long duration of the waves were observed for the relatively short-period pre-treatment of keratinase produced from the Bacillus sp. KN-517 stain. The applications of this keratinase to permanent wave considering the treatment time and temperature according to hair conditions would make consumers satisfied. The treatment preference of the keratinase of Bacillus sp. KN-517 confirmed for the possibility as hair cosmetics for was analyzed by using questions. For the custormers having thick hair or hairy hair, the keratinase treatment was applied as a pre-treatment to obtain a conclusion as follows. For the permanent using keratinase, the elasticity of the most curls was excellent for the 88% of the population. For the 25 customers with thick hair treated by kerastinase as a treatment source, the hair becomes glossy and light for the 92% of the population.
