Alginate is an anionic polymer, biocompatible, hydrophilic, and biodegradable under normal physiological conditions. Alginate materials are used to absorb wound fluid and to donate water into a dry wound, interacting between the alginate materials and wound surface to create a moist local environment. This study was developed the neo-alginate film by the casting evaporation method, which is a room temperature-dried gel and cross-linked with covalent bond. Through in vitro test, the alginate film morphology and cell viability were varified for required properties of wound dressing applications, as well as in vivo biocompatibility and wound healing ability with full thickness wound rat model. The results showed that alginate film had no cytotoxicity and healing effectivity in dermal wound treatment. The full-thickness skin defects were produced in the dorsal area of rats, and the skin wounds were covered with alginate. When alginate was used with occlusion the mean rate of healing was similar that seen under polyurethane film, and the epidermis was also at a mature stage of repair. The experiments on full thickness wounds confirmed earlier findings with alginates; they are efficient haemostatic agents that are well tolerated by the body fluids and cellular components. The results shown in a rat full-thickness excision wound model that alginate dressing can augment natural wound healing with protect dermal wound from bacterial contamination, prevent heat, protein and fluid losses, and provide a suitable environment for epithelialization. These findings suggest that topical alginate treatment for skin defects was very effective in healing of the wound. In conclusion, the alginate film showed excellent light transmittance, moisture retention and fluid drainage to promote healing ability compared with the gauze control. The results of the in vivo study indicated a promising application of the alginate film as an excellent moist wound dressing.
