Gastrointestinal mucosa is exposed to heavy load of commensal and pathogenic microorganisms but maintains the mucosal homeostasis through APCs containing dendritic cells (DCs) conditioned by mucosal environment. However, the balance is lost by infection of pathogen or an antigen influx. These counter events frequently occur in mucosal inductive site, Peyer’s patch (PP), since both commensal bacteria and luminal antigens are transported by M cells present in the site, although the factors regulating the balance are not well understood yet. In fact, M cells are known as specialized epithelial cells to take up the luminal antigens and microorganisms. This rapid and effective transcytotic activity of M cells makes them attractive target to mucosal vaccine, which required both the effective delivery of antigen and induction of antigen specific immune response. However, simply transport of antigen into the M cells does not guarantee the induction of antigen specific immune response. Therefore, first and second part in this study assumed that the apical receptor targeting of antigen in M cells could evoke antigen specific mucosal and systemic immune response. In order to describe this hypothesis, purpose of the first part is the identification of M cell targeting ligand, which could effectively transport the conjugated antigen into the M cells and induce the antigen specific immune response. To search for the M cell targeting ligands, in vitro M cell coculture model was established and confirmed by M cell specific antibody (NKM 16-2-4), GP2, and UEA-1; then ligands interacting with M-like cells were obtained from biopanning by phage display library. Among the ligands, it monitored that Co1 ligand not only specifically interacted with both mouse M cell and human M-like cell but also promoted the delivery of fused antigen into mouse PP. In addition, when Co1 ligand was applied into the oral vaccine model system, antigen specific immune response was enhanced in both systemic and mucosal compartment. Therefore, these results suggest that Co1 ligand is M cell targeting ligand, which could be used to promote antigen specific immune response in both the systemic and the mucosal compartment. It was thought that these results originated from interaction between specific receptor and Co1 ligand on M cells such as interaction with GP2 and FimH+ bacteria. Therefore, identification of apical receptor interacting with Co1 ligand on M cells is goal of part II. Interestingly, M cell targeting ligand, Co1 has 60% homology to OmpH α1 helix of Yersinia enterocolitica, which has been shown to share homology with that of the Skp α1 helix of E.coli, a chemotactic ligand of C5aR. It is plausible that M cell express C5aR. Accordingly, the second part of this study described that not only was identified the C5aR expression on M cells but also its exploiting by Y. enterocolitica was observed. Expression of C5aR in human M-like cells was confirmed by measuring the expression level of mRNA and protein and its apical expression was also monitored in mouse M cells. Furthermore, M cell targeting and induction of antigen specific immunity through C5aR were confirmed in oral vaccine model using C5aR knock-out mouse. Interestingly, during invasion of Y. enterocolitica, C5aR of mouse M cell was not only clustered but also phosphorylated nearby Y. enterocolitica. Moreover, C5aR on M cells could be exploited target of Y. enterocolitica through crosstalk pathway with TLR-2 because it is monitored that inhibition of C5aR by C5aR antagonist is caused in survival reduction of internalized Y. enterocolitica. Hence, C5aR is not only expressed, but also acts as target receptor on M cell. In that sense, PP are regarded as the “Achille’s heel” of mucosal barrier because of pathogens transpassing by M cells, but immune network for maintaining the homeostasis in PP was not well understood. Therefore, the third part of this study tried to identify the immune modulator in PP. Expression of human cathelicidin AMP, LL-37 was confirmed in human M-like cells as chicken M cells. In addition, it was found that LL-37 could regulate the expression of specific cytokine mRNA in PP lymphocytes.
