Rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 편백나무 잎 추출물의 세포막 보호 효과를 측정하였을 때 ethyl acetate분획과 aglycone 분획 모두 농도 의존적(5 ~ 25μg/mL)으로 광용혈 억제 효과를 나타내었다. In this study, evaluation of antioxidative activities and componential analysis of C. obtusa leaf extracts were carried out. Reactive oxygen species (ROS) scavenging activities (OSC50) of C. obtusa leaf extracts on ROS generated in Fe3+-EDTA/H2O2 system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction (0.22 μg/mL) and aglycone fraction of C. obtusa leaf extracts (0.20 μg/mL) showed more prominent ROS scavenging activity than L-ascorbic acid (1.50 μg/mL). The cellular protective effects of fractions of C. obtusa leaf extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction and aglycone fraction of C. obtusa leaf extracts suppressed photohemolysis in a concentration dependent manner (5 ∼ 25 µg/mL), and they exhibited better cellular protective effect than the lipid peroxidation chain blocker, (+)--Tocopherol (Vitamin E). The inhibitory effect of ethyl acetate fraction(74.43 μg/mL) and aglycone fraction(53.8 μg/mL) of C. obtusa extracts on tyrosinase were invetigated, too. The aglycone fraction showed 4 times higher tyrosinase inhibitory effect than arbutin(226.88 μg/mL) known as a strong whitening agent. The aglycone fraction of C. obtusa leaf extracts showed 3 bands in TLC chromatogram and 3 peaks in HPLC chromatogram (360 nm). Three compounds were identified as taxifolin, quercetin and kaempferol. We investigated the stability of cream containing 0.25 % C. obtusa ethyl acetate fraction. The pH, viscosity, and absorbance were measured under the 4 different temperatures (4, 25, 37 and 45 ℃) and under the sun light during the 12 weeks. The control cream without containing the extract did not show pH change under the different temperatures mentioned above. However, the pH of the cream with the extract was decreased 0.11 at 4 ℃ and 25 ℃. Under the 37 ℃, 45 ℃ and sun light condition, the pH was decreased 0.55, 0.84 and 0.59, respectively. After treating the cream for 12 weeks under the different temperatures, the viscosity was measured for the cream containing the extract and control cream. The values were decreased by 2,404 cPs in average compared to the initial value for the former and control cream were decreased by 1,296 cPs in average for the latter. On the other hand, the viscosity of control cream treated under the 45 ℃ for 12 weeks was decreased (1,915 cPs) relative to the cream containing the extract, which showed 3,810 cPs decrease in viscosity. The cream containing the extract did not show absorbance change at 4 ℃ ∼ 37 ℃ for 12 weeks. Instead, the absorbance of the cream treated under 45 ℃ and sun light condition was decreased 32.5 % and 35.2 %, respectively. This decrease in absorbance is relatively small compared to the 54.8 % decrease of the extract sampled from the cream using ethanol solution. This indicates that the extract is stabilized in the cream. In addition, any change in color or smell was not observed through 12 weeks of the experimental time period. Also physical changes as creaming and cohesion were not shown. The results show that the cream containing C. obtusa leaf extract was relatively stable. These results indicate that the fractions of C. obtusa leaf extracts can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging 1O2 and other ROS, and protect cellular membranes against reactive oxygen species. The fractions of C. obtusa leaf extracts can be applicable to new functional cosmetics for antioxidant and whitening effects.
