The present study investigated the protective effects of diallyl disulfide (DADS) against carbon tetrachloride (CCl4)-induced acute hepatotoxicity and oxidative stress in rat liver. We also investigated the effect of DADS on induction of apoptotic cell death and inflammatory response in hepatic tissue caused by CCl4 intoxication. The following five experimental groups were evaluated: (1) vehicle control, (2) DADS (100 mg/kg/day), (3) CCl4 (2 ml/kg/day), (4) CCl4&DADS 50 (50 mg/kg/day), and (5) CCl4&DADS 100 (100 mg/kg/day). DADS at 50 and 100 mg/kg was administered orally once daily for 5 consecutive days, prior to CCl4 administration. CCl4 caused a significant decrease in hepatic cytochrome P450 2E1 (CYP2E1) expression and severe hepatotoxicity as evidenced by a significant elevation in serum aspartate aminotransferase and alanine aminotransferase levels, increased hepatic cell apoptosis, Bcl-2-associated X (Bax) protein, cytochrome c, cleaved caspase-3 protein expression, and caspase-3 immunopositivity. Additionally, CCl4 treatment caused an increase in levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (Cox-2) mRNA and protein expression in hepatic tissues. The significant decreases in reduced glutathione content, catalase, superoxide dismutase, glutathione peroxidase, and glutathione reductase activities and the increase in malondialdehyde concentration indicated that CCl4-induced hepatic injury was mediated through oxidative stress. In contrast, DADS pretreatment significantly attenuated oxidative stress and the hepatotoxic effects caused by CCl4 administration. Moreover, DADS effectively suppresses the induction of inflammatory mediators, iNOS and Cox-2, attenuates hepatic cell apoptosis, and decreases the Bax, cytochrome c, and cleaved caspase-3 expression caused by CCl4 administration. These results indicate that DADS prevents hepatotoxicity induced by CCl4 in rats and that protective effects of DADS may be due to its ability to decrease the metabolic activation of CCl4 by inhibiting CYP2E1, subsequently preventing CYP2E1 degradation or inactivation, and its potent antioxidant activity and anti-inflammatory activities in rat liver.
