연구목적: 본 연구에서는 나복자의 물 전탕 추출물이 간세포의 지방독성(Lipotoxicity)에 미치는 효과를 연구하기 위하여 방법: HepG2 cell에서 palmitic acid에 의한 지방독성을 일으켜 상기 약물들에 대한 cell viability를 조사하였고, 나복자의 물 전탕 추출물이 reactive oxygen species 생성에 미치는 영향, ER 스트레스 마크인 PERK, IRE1α, CHOP, ER 내 thiol의 양에 미치는 효과를 조사하여 다음과 같은 결론을 얻었다. 1. 나복자의 물 전탕 추출물은 palmitic acid로 유발한 지방독성에 의해 감소된 cell viability를 증가시킨다. 2. 나복자의 물 전탕 추출물은 palmitic acid에 의해 증가된 reactive oxygen species 생성을 억제한다. 3. 나복자의 물 전탕 추출물은 palmitic acid로 인해 증가된 PERK, IRE1α,의 발현을 증가시키고 CHOP의 발현증가에는 효과가 없다. 4. 나복자의 물 전탕 추출물은 palmitic acid에 의해 감소된 ER의 thiol을 증가시킨다. 이상과 같이 나복자의 물 전탕 추출물은 간지방의 세포모델인 human hepatoma HepG2 세포주에 palmitic acid를 처리한 지방독성 모델에서 세포 보호효과와 reactive oxygen species 생성을 억제하는 효과가 있음이 관찰되었고 ER에 야기된 단백질 미접힘 반응을 억제하고 악화된 산화적 환경을 개선하는 효과가 있어 나복자는 비알코올성 지방간 질환(Nonalcoholic fatty liver disease, NAFLD)의 치료에 임상적 의의가 있을 것이라 기대된다.

Objective : The aim of this study was to investigate the mechanism of protective effect of the Raphanus sativus Linne (RSL) water extract on palmitic acid-induced lipotoxicity. Methods : In order to induce lipotoxicity on HepG2 cells, the cells were incubated with the water extract of RSL and then the media was replaced with a 0.7 mM PA-containg media with 4 % of BSA. Cell viability was measured by MTT assay and reactive oxygen species or the thiols in ER were evaluated by fluorometer or confocal microscopy. And western blot was performed to evaluate the expression of PERK, IRE1α, and CHOP. Results : The following results are obtained. 1. The RSL increased the cell viability of the HepG2 cells under the condition of the PA-induced lipotoxicity. 2. The RSL decreased the ROS level of the HepG2 cells under the condition of the PA-induced lipotoxicity. 3. The RSL decreased the expression level of PERK and IRE1α in the HepG2 cells under the condition of the PA-induced lipotoxicity. However, it has no significant effect on the CHOP level of the cells under the corresponding condition. 4. The RSL increased the thiol level in the ER of the HepG2 cells which was decresed by the PA-induced lipotoxicity. Conclusion : These results indicate that Raphanus sativus Linne (RSL) water extract has a protective effect of cell viability of palmitic aicd-treated HelpG 2 cell line and reduced ROS level. It also reduced unfolded protein response and oxidized balnce in the ER of the cells induced by palmitic acid. Therefore, RSL may contribute to the clinical treatment of nonalcoholic fatty liver disease (NAFLD).
