HPLC 분석 결과에 따르면, 우방자추출발효물의 β-glucosidase 활성은 리간드 성분의 악틴 배당체로를 악티게닌 성분으로 전환시켰으며, 배양 전 우방자추출물에 다량 함유된 카페익산(caffeic acid) 배당체도 단일성분의 카페익산으로 전환되었다. 우방자추출물은 발효 후 향상된 항산화 및 염증 완화 효과를 보였다. 이러한 효과는 발효 배양물 내 악틴, 악티게닌 그리고 카페익산 이외의 다른 대사물질과의 시너지에 의한 결과로 생각되어 향후 실험은 우방자추출발효물로 진행하였다. 우방자추출발효물은 200 ㎍/㎖ 이하 농도에서는 세포독성을 전혀 보이지 않았으며, 50 ㎍/㎖에서 21% 정도의 세포증식 효과를 보였다. 자외선 조사 조건에서 세포내 항산화효과가 우수하게 나타났으며, 콜라겐합성 촉진효과도 50 ㎍/㎖ 농도에서 51 %로 우수하게 나타났다. 우방자추출발효물은 라미닌 합성에 대해서도 비슷한 효과를 보였다. 콜라겐합성 경로에는 Smad 2의 인산화가 관여한다. 상처치유효과실험 결과 대조군에 비해 우방자추출발효물 처리군에서 세포의 이동이 향상되었다. 우방자추출발효물은 자외선 조사에 의해 유도되는 염증관련 사이토카인의 생성과 세포독성을 농도의존적으로 억제하였다. 자외선에 의해 과생성된 MMP-1(collagenase)의 발현도 우방자추출발효물에 의해 현저히 감소하였다. 우방자추출발효물은 세포 노화에 관련된 지표인 β-galactosidase의 발현도 효과적으로 억제하였다. 피부장벽기능을 나타내는 각질분화 지표 단백질인 filaggrin, involucrin, loricrin의 발현량을 측정한 결과, 우방자추출발효물은 이들의 발현을 2-D와 3-D 배양 모두에서 증가시켰다. 최종적으로 인체 피부에 대한 실험에서 우방자추출발효물 3%를 함유한 화장품을 피부에 바른 군에서는 대조군에 비해 피부 보습 및 주름개선 효과가 유의하게 나타났다. 결론: 우방자추출발효물은 자외선에 대한 광방어효과와 세포보호효과가 우수하기 때문에 향후 광방어 및 항노화 효과를 보이는 화장품의 신소재로 활용 가능성이 높다.

Purpose: The human skin is constantly exposed to environmental irritants such as ultraviolet (UV), smoke, chemicals. Free radicals and reactive oxygen species (ROS) generated by these environmental factors play critical roles in cellular damage. Exposure to solar radiation, particularly its UV component, induces a variety of harmful effects on human health. Some of these effects include sunburn cell formation, photoaging of the skin and carcinogenesis. The purpose of this study was to investigate the photo-protective effect of fermented Fructus arctii extract (G-FAE) on the UV-irradiated human dermal fibroblasts (HDFs) and HaCaT keratinocytes, and anti-aging effect of G-FAE on human skin. Materials and methods: Fructus arctii extracts (FAE) containing diverse glycosides were cultured with Grifola frondosa HB0071 mycelia producing β-glucosidase. Fermented FAE (G-FAE) compounds were subjected to HPLC analysis. Intracellular ROS production was measured by DCF fluorescence upon exposure to UVB 20 mJ/cm2. The expression of collagen and laminin was monitored using Western blot analysis or enzyme immunoassay (EIA). The wound healing assay was performed by measuring cell migration after wound scratch in human dermal fibroblasts (HDFs). Anti-inflammatory effect was monitored by quantifying secreted interleukin 1α (IL-1α) and prostaglandin E2 (PGE2) after UVB irradiation in HaCaT keratinocytes. To assess the expression level of matrix metalloproteinase-1 (MMP-1), enzyme linked immunosorbent assay (ELISA) and RT-PCR analysis were conducted in UVA-irradiated HDFs. Skin barrier function were tested by monitoring the expression of key protein markers for epidermal differentiation, filaggrin, involucrin and loricrin, in a 2D and a 3D culture of HaCaT keratinocytes. For testing anti-aging and moisturizing effects of G-FAE on human skin, Skin Visiometer and Corneometer were used. Results: β-glucosidase activity reached a peak of 85.9 mU/ml at 72 hours during the fermentation of FAE. Accordingly, the levels of arctigenin and caffeic acid increased in the G-FAE, which showed higher antioxidant and anti-inflammatory activities than those of FAE as assessed by NBT, DPPH, and 5-lipoxygenase assays. G-FAE also showed a potent free radical scavenging activity in UVB-irradiated HDFs. Moreover, G-FAE had a marked stimulatory activity of collagen biosynthesis. Treatment of HaCaT keratinocytes with G-FAE resulted in the increased expression of laminin-5, a key basement membrane component. G-FAE increased the cell migration over the wounded monolayer, suggesting that it has beneficial effect on wound healing. G-FAE decreased expression of a proinflammatory cytokine IL-1α about 30% at 50 μg/ml in HaCaT keratinocytes. G-FAE also decreased UVB irradiation-induced production of PGE2 by HaCaT keratinocytes in a dose dependent manner (p < 0.05). It is well established that UVA irradiation elevates the expression level of MMP-1. G-FAE reduced the levels of both MMP-1 mRNA and protein in UVA-irradiated HDFs. Treatment of HaCaT keratinocytes with G-FAE in vitro increased the expression of key protein markers for epidermal differentiation such as filaggrin, involucrin and loricrin.
