Through the analysis of microsolvating effect on the isomers’ relative stability for glycine, alanine, and valine with the hydrophobic side chains, it is found that the microsolvating properties of amino acids strongly depend on the size and the properties (hydrophilicity/hydrophobicity, basicity). For the amino acids with hydrophobic side chains (glycine, alanine, valine), their zwitterions are stabilized by 5~7 water molecules, but as of the size of the side chain increases by fewer water molecules. On the other hand, the zwitterionic forms of the amino acids (arginine and lysine) with strongly basic side chains are stabilized even by 1~3 water molecules. The cyclic amino acid proline is predicted to exist as a zwitterion under the influence of 4 water molecules, presumably due to the stronger basicity of the secondary amine in it. Lastly, for the dipeptide diglycine, we predict that much more (>8) water molecules may stabilize the zwitterion, because of the presence of the peptide bond and the greater distance between the carboxyl and amino group. The mechanism of SN2 reactions are usually depicted by an oversimplified scheme, omitting the effects of the counterions and solvents. This thesis presents the new type of SN2 reactions, in which the bulky alcohols, oligoethylene glycols and ionic liquids increase the reaction rates acting on the counterions as Lewis bases. The nucleophiles may react as ion pairs or as naked nucleophiles. This type of organocatalysis is analyzed in detail by examining the structures of the pre-reaction complexes and the transition states, in comparison with experimental results. The effects (TriEG < TetraEG < PentaEG) of the length of the side chains in the organocatalysis by n-oligoethylene glycols (OligoEGs) are interpreted by increasing number of oxygen atoms stabilizing the metal cation. The better efficiency of Cs+ than K+ is the smaller Coulomb influence of Cs+ on the nucleophile. As for the reactivity of different nucleophiles, the difference in the ion binding strengths with the metal cation seems to by the major factor. The synergetic promotion of reaction rares by the OligoEGs–ionic liquid complexes are also examined. The organocatalysis by [monohexaEGmim][OMs] proceeds by a mechanism (ion-pair SN2 reaction) similar to that by OligoEGs, but [dihexaEGim][OMs] seems to “dissociate” the ion pair, producing the “naked” nucleophile by the combined effects of the OligoEGs and the ionic liquid moieties.
