Lung cancer is the leading cause of cancer deaths. The 5-year survival rate of lung cancer is very low because of late diagnosis resulting from undetectable symptoms. Although deregulation of several transcription factors have been reported to contribute to the pathogenesis of lung cancer, associated network is not fully understood. In this thesis, I focused on the construction of transcriptional regulatory network of transcription factor Forkhead box A2 (FOXA2) which functions as tumor suppressor in variety types of cancers. I identified four novel interacting partners, histone deacetylase 1 (HDAC1), histone deacetylase 3 (HDAC3), heat shock protein 90kDa alpha (cytosolic), class A member 1 (HSP90A) and heat shock 70kDa protein 1A (HSPA1A) using proteomic approach and experimental approach and four novel target genes, cyclin-dependent kinase inhibitor 1A (p21), BCL2-associated X protein (BAX), nuclear receptor subfamily 0, group B, member 2 (NR0B2), cell adhesion molecule 1 (CADM1) using bioinformatics approach and experimental approach. These novel interacting partners of FOXA2 physically interacted with FOXA2 and the mRNA level of novel target genes were upregulated in FOXA2 overexpressed H1299 cells. Especially, FOXA2 activated transcription of p21 and BAX via direct binding to each promoter in vivo and in vitro in H1299 cells. A newly identified inducers for FOXA2, trichostatin A (TSA) upregulated p21 and BAX expression through induction of FOXA2 protein, which was confirmed by knockdown of FOXA2 resulting in inhibition of TSA-induced expression of p21 and BAX. Induction of FOXA2 protein expression by TSA treatment was confirmed in time and dose dependent manner in H1299 cells. Regulation of TSA-induced FOXA2 expression was controlled at the translational level. The supporting evidences were that no alteration of FOXA2 mRNA expression was observed by TSA treatment in H1299 cells, even though its detectable level at the basal condition. Additionally, translation inhibitor cycloheximide treatment repressed TSA-induced FOXA2 expression. Increase of FOXA2 expression by TSA treatment was regulated by phosphorylated eukaryotic translation initiation factor 4E binding protein 1 (4EBP1). Phosphorylation of 4EBP1 was increased in TSA-treated H1299 cells coincided with the increased expression of FOXA2 protein, which was supported by inhibition of 4EBP1 phosphorylation by rapamycin leading to repression of TSA-induced FOXA2 expression. Furthermore, ribonucleoprotein immunoprecipitation (RNA-IP) assay revealed that association of 4EBP1 with FOXA2 mRNA was decreased through phosphorylation of 4EBP1 by TSA treatment. In addition, although mammalian target of rapamycin (MTOR) and regulatory associated protein of MTOR, complex 1 (RPTOR), upstream regulator of 4EBP1, interacted with histone deacetylase 2 (HDAC2) in the basal condition, these interactions were disassociated by TSA treatment. Thus, binding of TSA to active motif of HDAC2 may contribute to phosphorylation of 4EBP1 by releasing MTOR and RPTOR for activation, resulting in induction of FOXA2 expression. Taken all together, mapping of regulatory network of FOXA2 in lung cancer is essential for precisely controlling tumor development and progression. I additionally suggest that full understanding of transcriptional regulatory network of FOXA2 is important for the future therapeutic application for lung cancer which can be extended to other cancers.
