농작물을 해하는 해충을 방제하기 위해 많이 사용되는 carbamate계 살충제는 독성과 잔류성이 문제되고 있어 효율적인 제거기술도 필요한 실정이다. 환경 중 carbamate계 살충제와 그 대사체의 동태를 구명하는 것은 발생할 수 있는 위해성 문제를 감소시키기 위해 중요한 일이기 때문에 본 연구에서는 가장 많이 사용되고 있는 carbamate계 살충제인 carbaryl 및 carbofuran을 대상으로 peroxidase, zerovalent ion (ZVI) 및 활성탄 같은 다양한 촉매제를 처리하여 수중에서의 두 농약의 제거 정도를 조사하였다. 수중에서 carbaryl 및 carbofuran의 잔류량은 horseradish peroxidase(HRP)를 처리하였을 때 거의 변화 없었다. 그러나 carbaryl의 대사체인 1-naphthol은 HRP와 빠르게 반응하였다. 또한 catechol의 존재 하에서 HRP에 의한 1-naphthol의 반응산물이 더 많아지는 것을 확인하였다. Catechol을 첨가하지 않았을 때 1-naphthol의 반응산물이 오직 7%만이 생성된 것에 비해 0.3 mM H2O2를 포함하는 0.1 M의 sodium phosphatebuffer 용액(pH 7.0) 중에서 3.0 mM catechol과 0.5 unit/mL HRP를 첨가한 후 3시간을 반응시켰을 때 1-naphthol의 반응 생성물은 79%까지 증가하였다. 용액의 pH와 효소활성이 증가할수록 1-naphthol 반응산물의 생성율 역시 증가하였으며 catechol의 반응산물을 더 확인하기 위하여 high performance liquid chromatography(HPLC)와 mass spectrometry(MS)를 이용한 반응생성물을 조사하였다. 반응생성물은 1-naphthol autoxidation 생성물, 1-naphthol과 그 autoxidation 생성물의 결합 생성물 그리고 1-naphthol oligomer로 세 가지 형태가 확인되었으며 liquid chromatogram-tandem mass spectrometry (LC-MS/MS)에서 191 m/z의 분자량을 가진 5,8-dihydroxy,1,4-naphtholquinone 물질이 확인되었다. 이러한 결과와 앞선 보고들을 바탕으로 catechol의 존재 하에 HRP에 의한 1-naphthol 반응산물의 생성 경로를 제안하였다. Carbofuran과 carbaryl의 제거 시험을 ZVI가 단독으로 있을 때와 다른 철염이 공존할 때를 대상으로 수행하였다. 두 농약 모두 ZVI가 단독으로 있을 때보다 철염과 공존할 때 분해 효율이 증가되었다. 즉, ferric염이 ferrous염보다 수중에서 carbofuran의 제거에 더 효율적이었으며 ferric nitrate는 ZVI가 있을 때 가장 우수한 carbofuran 분해 효율을 나타내었다. 철염의 효율성 순서는 Fe(NO3)3>FeCl3>Fe2(SO4)3>FeCl2>FeSO4이었다. 이 반응으로부터 생성된 carbofuran의 3가지 분해산물을 GC-MS를 이용하여 확인한 결과 2,2-dimethyl-2,3-dihydro-benzofuran-3,7-diol (180 m/z), 7-hydroxy-2,2-dimethyl benzofuran -3-one (178 m/z) 및 2,2-dimethyl-2,3-dihydro-benzofuran-7-ol (164 m/z)으로 밝혀졌다. 세 가지 종류의 탄소화합물Black Pearl 2000 (BP 2000), extruded activated carbon (EAC) 및 granular activated carbon (GAC)을 이용한 carbofuran 제거 능력이 평가되었다. 표면특성을 알고 있는 흡착제가 carbofuran을 어느 정도 흡착 제거하는지 초기 농도가 50-200 mg/L와 pH 2.0-8.0 사이에서 수행되었다. Carbofuran의 농도가 증가될수록 흡착 정도도 증가하였으며 초기 농도 200 mg/L에서 BP 2000에 대해 555 mg/g까지 최대 흡착량을 보였다. Carbofuran의 제거에 대한 흡착제의 효율성은 BP 2000>GAC>EAC의 순이었으며 평형시간에 흡착된 carbofuran의 양을 조사하여 Langmuir, Freundlich 및 Temkin 흡착등온선에 적용하였다. 그 결과 Langmuir 등온선이 가장 적합한 model이었으며 수 중 carbaryl의 제거에도 역시 세 가지 흡착제 모두 효과적인 것으로 나타났다. 따라서 높은 표면적을 가진 BP 2000, EAC 및 GAC는 수 중 오염된 carbaryl 및 carbofuran의 제거를 위해 흡착제로 사용될 수 있는 잠재성이 있었다. 이러한 연구결과들은 carbamate계 살충제인 carbaryl 및 carbofuran과 carbaryl의 대사산물인 1-naphthol을 수중 제거시키는데 HRP, ZVI 및 활성탄이 사용될 수 있음을 보여준다.

Carbamate insecticides are used to control insects in agricultural fields, but toxic and persistent, which need effective remediation technologies. The knowledge related to the behavior and fate of carbamate insecticides and their metabolites in the environment are important resources to reduce the risk accompanied with them. In this study, two popular and widely used carbamate insecticides, carbaryl and carbofuran, were treated using various catalysts, peroxidase, zerovalent iron (Fe0; ZVI), and activated carbon, to know their impacts in water. We found minor or no change when carbaryl and carbofuran were treated with horseradish peroxidase (HRP). However, the carbaryl metabolite 1-naphthol was readily transformed due to peroxidase effect in solution. When 1-naphthol transformation by peroxidase was occurred in the presence of catechol, significant enhancement of 1-naphthol transformation was obtained over that in the system without catechol. Result shows that, without catechol, 1-naphthol transformation was only 7%, but this reached to 79% because of its presence (3.0 mM) by peroxidase (0.5 unit/ml) after 3.0 h (pH 7.0), in the solution of 0.1 M sodium phosphate buffer containing 0.3 mM H2O2. Due to pH and enzyme activity increase in solution, 1-naphthol transformation rate was also accelerated. In order to further confirmation of catechol effect, the products formed were characterized using high performance liquid chromatography (HPLC) and mass spectrometry (MS) analysis. Three types of reaction products were identified, such as 1-naphthol autoxidation products, 1-naphthol and 1-naphthol autoxidation coupling products and 1-naphthol oligomers. The formation of molecular ion m/z 191 was selected for fragmentation in liquid chromatography-tandem mass spectrometry (LC-MS/MS) and confirmed as 5,8-dihydroxy,1,4-naphthoquinone. Based on the above observations and previous reports, we proposed a route for the transformations of 1-naphthol by peroxidase in the presence of catechol. The removal of carbofuran and carbaryl were carried out using zerovalent iron ZVI alone and ZVI in the presence of different iron salts. The degradation rate and degradation efficiency were also studied. In all cases, the degradation rate and efficiency were increased in the presence of iron salts by ZVI rather than ZVI alone. The results obtained indicated that ferric salts could more efficiently remove carbofuran from solution than ferrous salts, and ferric nitrate was found to be most effective iron salt for degradation of carbofuran in aqueous solution in the ZVI presence. The order of effectiveness of iron salts were found to be, Fe(NO3)3FeCl3Fe2(SO4)3FeCl2FeSO4. Three degradation products of carbofuran were identified by GC-MS analysis, such as 2,2-dimethyl-2,3-dihydro-benzofuran-3,7-diol (180 m/z), 7-hydroxy-2,2-dimethyl-benzofuran-3-one (178 m/z), and 2,2-dimthyl-2,3-dihydro- benzofuran-7-ol (164 m/z). The impact of three commercial carbons, Black Pearl 2000 (BP 2000), extruded activated carbon (EAC) and granular activated carbon (GAC), on carbofuran removal was evaluated. The adsorbents surface properties were characterized. Batch experiments were conducted to show the effects as a function of pesticide initial concentration and pH over the range of 50.0 to 200.0 mg/L and 2.0 to 8.0, respectively. The equilibrium adsorption for all cases increased with increasing initial carbofuran concentration and the maximum adsorption capacity was obtained for BP 2000 up to 555.55 mg/g at 200.0 mg/L of initial concentration. The removal potential was found in order of BP 2000GACEAC. The amount of carbofuran adsorbed at equilibrium was applied into Langmuir, Freundlich and Temkin isotherm models. The Langmuir isotherm model was fitted best. BP 2000, EAC and GAC also effectively removed carbaryl from water. Thus, BP 2000, EAC and GAC having high surface area can be used as a potential adsorbent to remove carbaryl and carbofuran from contaminated water. This research effort provides a useful means in remediation of pesticides carbaryl and carbofuran, and carbaryl metabolite, 1-naphthol.
