The Chaenomeles sinensis fruit is commonly used to treat conditions such as pneumonia and bronchitis, but a dearth of research on the leaf exists. In this study, we investigated the anti-inflammatory effects of C. sinensis leaf extract (CSL) on LPS-stimulated RAW 264.7 cells. The 70 % ethanol extract of CSL (CSLE) significantly inhibited NO, IL-6, and TNF-α production; additionally, the CSLE suppressed LPS-stimulated inducible NOS (iNOS), IL-1β, and phospho-STAT1 expression. In this study, we investigated whether CSLE decreases the production of inflammatory mediators through the inhibition of the TRIF-dependent pathways. Our results suggest that CSLE attenuates LPS-stimulated inflammatory responses in macrophages via the regulation of the TLR4 pathways, providing scientific support for its traditional use in the treatment of a variety of inflammatory diseases.
