실험에 사용된 병원성 미생물은 E. coli, S. aureus, P. citrinum, A. niger 총 4종 이었다.

The tea drink products currently distributed in the market contain no preservatives due to the well-being trend in most cases. However, They possesses a risk of microbial contamination and proliferation as they have weak conservative properties with a neutral pH and are distributed at room temperature. Since synthetic preservatives have been subject to constant safety controversies and received poorly by consumers, many manufacturers use natural preservatives. It is, however, limited to use natural preservatives and difficult to apply them to products owing to their high prices. This study thus selected thyme oil, tea tree oil, orange oil, and grapefruit extract, which were considered to have antibacterial activity and sensory fitness for tea drinks among the natural essential oils capable of replacing synthetic preservatives, and compared them with sodium benzoate. The investigator implemented measurement with paper disc and MIC methods and carried out a sensory evaluation by selecting natural essential oils with superior antibacterial activity and applying them to tea drinks for optimizing concentration. Totally four kinds of pathogenic microorganisms were used in the experiment including E. coli, S. aureus, P. citrinum, and A. niger. Tea tree oil began to show superior antibacterial activity to the other natural essential oils at the concentration of 20%, and thyme oil showed superior antibacterial activity to the other natural essential oils in A. niger with 18 mm of bacteria growth in A. niger at the concentration of 20% and no more growth of bacteria in P. citrinum at the concentration of 40% or higher. The results of antibacterial activity among the natural essential oils based on the MIC method were similar to those of the paper disc method with thyme oil demonstrating the best antibacterial activity. It showed a range of 0.07~90%, 0.05~96%, 0.05~91%, 0.05~91% in antibacterial activities against E. coil, S. aureus, A. niger, P. citrinum, respectively. The investigator applied thyme oil whose superior antibacterial activity was demonstrated in the experiment to tea drinks and compared it with sodium benzoate. As a result, it showed similar antibacterial activity affect to those of sodium benzoate at the concentration of 0.03% or higher and better antibacterial effects in A. niger. Since the strong fragrance of thymol, one of the core ingredients of thyme oil, lowered the sensory quality of a product with the increasing content of thyme oil and accordingly rising antibacterial strength, the investigator applied it to tea drinks and conducted a sensory evaluation to find optimum amounts of usage. These results indicate that grain tea products had differences in taste and odor from the old ones and were thus not in harmony with thyme oil. Herbal tea products, on the other hand, had few differences from the old ones, which means that thyme oil was effective to red ginseng and ginseng tea products. Natural essential oils have their unique tastes and odors due to their principal ingredients, thus having influences on quality changes according to the product types.
