Hibiscus. syriacus L. which is found in Asia, is an important plant ornamental shrub. It has been widely cultivated as a decorative shrub because its flower is national flower of Korea. Hibiscus syriacus L. has been used to Vaginosis, shigellosis, hemorrhoids, anthelmintic, antipyretic, eczema, psoriasis in Asian traditional medicine. However, research on keratinocytes is inadequate. Hence, we identified the composition of absolute from Hibiscus syriacus L. flower absolute (HSF absolute) using gas chromatography-mass spectrometry analysis. We also examined the effect of HSF absolute in HaCaT cells using XTT assay, Boyden chamber assay, collagen synthesis measurement, ex vivo sprout-out growth assay, western blot, and in vivo rat tail wound healing assay respectively. 10 major active compounds palmitic acid (18.94%), linoleic acid (8.57%), stearic acid (5.87%), 9-nonadecene (5.82%), cycloartenol (3.31%), γ-sitosterol (2.22%), lupenyl acetate (1.52%), myristic acid (1.16%), squalene (0.83%) and γ-tocopherol (0.81%) were identified from HSF absolute. We determined the effect of HSF absolute 0.1 mg/mL to proliferation (133.05 ± 2.71%), migration (189.08 ± 4.34%) and sprout-out growth (458.0 ± 6.38%) rates of keratinocytes. In addition, HSF absolute 0.1 mg/mL also enhanced increase phosphorylation of the Erk 1/2 (334.12 ± 3.31%) and Akt (403.16 ± 3.66%) and induced the synthesis of type I (143.61 ± 1.78%) and IV (150.43 ± 1.82%) collagen in HaCaT cells. Moreover, promoted wound closure in the dorsal side skin of rat tail. As shown by XTT assay, Boyden chamber assay, ex vivo sprout-out growth assay and western blot results HSF absolute exhibited similar effects of EGF on proliferation and migration patterns of HaCaT cells. These results of in vitro, ex vivo and in vivo studies imply that HSF absolute induced the cell proliferation, migration, synthesis of type I, IV collagen and sprout-out growth of HaCaT cells. through Erk 1/2 and Akt phosphorylation and rat tail wound healing. Therefore, this study suggests that HSF absolute may show promise as a good material for use in cosmetics and alternative medicine.
