본 연구는 유산균의 면역증진 효과를 유세포 형광분석으로 확인하였고, 유산균의 황색포도상구균항균활성을 실험하여 항아토피 기능성이 있는 유산균을 선발하고 그 배양방법을 검토하였다. T cell에서 CD4+CD25+foxp3+ 증가는 Lb. plantarum > Lc. lactis의 순으로 면역증진효과를 보였고, 황색포도상구균에 대한 항균력은 Lc. lactis, Lb. plantarum의 순으로 나타냈다. 유산균의 혼합배양방법은 Lc. lactis 배양액을 첨가한 배지에 Lb. plantarum을 배양한 실험군에서는 항균활성의 증가가 없었으나 Lb. plantarum 배양액을 첨가한 배지에 Lc. lactis를 배양한 실험군에서 황색포도상구균의 항균활성 증대를 확인하였다. 항균력을 증가시키는 유산균 첨가배지법의 최적조건은 Lb. plantarum 배양상층액을 10% 첨가한 배지에서 Lc. lactis 배양으로 나타났다. 이때 유산균 Lc. lactis 첨가배지법에서 황색포도상구균의 증식 억제력이 순수배양에 비해 1.29배 높아졌다.

본 연구는 RBL-2H3 비만세포를 이용하여 유산균의 첨가 배양방법으로 배양한 유산균이 아토피 피부염의 원인이 되는 알레르기 반응에서 얼마나 억제능을 갖는지 알아보았다. 유산균이 GATA-1, GATA-2, NF-AT1, NF-AT2, NF-κB p65 전사인자들의 활성을 억제하는지를 분석하였고, 활성화된 비만세포에서 발현하는 cytokine IL-4와 IL-13을 조절할 수 있는지를 실험적으로 규명하였다. 그 결과 Lb. plantarum 배양상층액 첨가 Lc. lactis 배양액 > Lc. lactis > Lb. plantarum 배양액 첨가 Lc. lactis 배양액 > Lb. plantarum의 순으로 나타났다. 따라서 in vitro에서 Lb. plantarum 배양상층액을 10% 첨가한 배지에서 Lc. lactis 유산균을 배양한 첨가배지법이 활성화된 비만세포의 알레르기 반응으로의 분화 및 활성을 가장 잘 억제하고 관련 cytokine IL-4와 IL-13 유전자 발현을 선택적으로 조절하는 항알레르기 효과를 나타내었다.
이상의 결과를 바탕으로 유산균의 혼합배양 방법이 아토피 피부염을 효과적으로 억제하는지 임상실험 하였다. NC/Nag 생쥐에 유산균과 그 혼합배양방법을 경구투여하여 아토피 피부염 억제를 확인하였다. 혈액검사, IgE 측정, histology 분석, ALN 및 등 피부조직에서 세포분석, 유전자 검사, 비장세포에서 cytokine 측정 등을 통해 아토피 피부염 억제 효과를 실험적으로 규명하였다. 본 연구 결과를 종합하여 볼 때, Lb. plantarum 배양상층액 첨가 Lc. lactis 배양방법과 Lc. lactis 유산균에서 유의성 있는 효과를 보였다. 이로써 유산균과 유산균의 혼합배양방법이 면역증진, 황색포도상구균 항균효과, 비만세포의 알레르기 억제, 프로바이오틱스로서 복합 섭취했을 때 아토피 피부염의 억제에 더욱 효과적으로 적용되어 아토피 피부염의 면역 조절 효과로서 그 치료를 기대한다.

We conducted a screening and checked cultivation methods of lactic acid bacteria which has anti-atopic dermatitis function by identifying lactic acid bacteria's immune enhancement by FACS and antimicrobial activity against Staphylococcus aureus (S. aureus). The increased CD4+CD25+foxp3+ in T cell was result immune enhancement effect in the order of Lactobacillus plantarum (Lb. plantarum), Lactococcus lactis subsp. lactis (Lc. lactis), antimicrobial activity against S. aureus was result in the order of Lc. lactis, Lb. plantarum. Antimicrobial activity in Lb. plantarum culture with medium containing Lc. lactis culture broth was not enhanced, but antimicrobial activity of Lc. lactis culture with medium containing Lb. plantarum culture broth was enhanced. Optimum culture conditions for enhanced antimicrobial activity was Lc. lactis culture with medium containing 10% of heat-killed Lb. plantarum culture broth. In this condition, the anti-bacterial activity of Lc. lactis pure culture increased sharply at the end of log phase than poetry, restraint in the growth of S. aureus has increased 1.29 times.
We analyzed whether lactic acid bacteria could control the expression of IL-4 and IL-13 in activated mast cells and whether these bacteria could inhibit the activity of transcription factors such as GATA-1, GATA-2, NF-AT1, NF-AT2, and NF-κB p65. We characterized the effect of lactic acid bacteria cultured by this double-culture method on inhibition of allergic inflammatory reactions of RBL-2H3 mast cells, a cellular model of atopic dermatitis. The strongest anti-allergic effects of the lactic acid bacteria were seen in the following order: Lc. lactis broth cultured with medium containing Lb. plantarum culture supernatant > Lc. lactis > Lc. lactis broth cultured with medium containing Lb. plantarum culture supernatant > Lb. plantarum. Thus, Lc. lactis cultured in medium containing Lb. plantarum culture supernatant had the strongest inhibitory effect on the differentiation of mast cells during allergic reactions, which may be mediated through the selective regulation of expression of relevant genes.
As the animal test model for atopic dermatitis, NC/Nga mice are widely known. We examined comparison of skin manifestation and clinical skin features and severity of skin change, hematology, serum IgE elevation, histological features and status of dorsal skin, total cell number of dorsal skin and ALN, CD4+ and CD11b+Gr-1+ changes of total absolute numbers, eotaxin, and CCR3 mRNA expression, CD4+ & CD8+, B220+CD23+ and CD3+CD69+ changes of total absolute numbers, IL-5, and IL-13 mRNA expression, IL-4, IFN-γ, IL-5 and IL-13 level in NC/Nga mice induced by DNCB. From the above results, we concluded the cultivation methods of lactic acid bacteria which has anti-atopic dermatitis function that Lc. lactis broth cultured with medium containing Lb. plantarum culture supernatant.
