미세조류의 광합성 반응에서 생산된 산소를 하수처리에 이용하는 것은 송풍 에너지를 절감하는 새로운 대안으로 기대되고 있다. 그러나 활성슬러지 공법에 비해 상대적으로 긴 하수처리 시간, 낮은 적용 수심, 기상 여건에 따라 변동되는 처리수질 등은 기술 보급을 어렵게 하는 주요한 요인이다. 본 연구에서는 미세조류-박테리아 공배양 시스템의 광합성과 산소 생산 특성, 하수처리 시간 단축을 위한 새로운 방안에 대한 연구를 진행하였다. 먼저, 미세조류-박테리아 공배양 바이오매스의 광합성 산소 생산 및 하수처리 특성을 파악하기 위한 기초연구로서 청색, 녹색, 적색 및 무색 LED 조명 조건에서 광합성 및 하수처리 특성을 파악하였다(제 4 장 및 제 5 장). 이러한 기초 연구를 바탕으로 미세조류-박테리아 공배양 시스템에서 질산화 반응을 촉진하기 위한 대안으로 유동 담체의 적용 가능성을 평가하였다. 즉, 광생물 반응기에 유동 담체를 충진하고 유기물, 영양 염류, 침전성 등에 미치는 영향을 파악하고 하수처리 시간 단축 효과를 검토하였다(제 6 장). 제 7 장에서는 제 6 장에서의 유동 담체의 하수처리 특성에 대한 이해를 바탕으로 미세조류-박테리아 공배양 시스템의 하수처리 시간 단축 방안을 모색하였다. 즉, 광생물 반응기에 유동 담체를 충진하여 질산화 반응을 촉진하고, 종속영양 미생물에 의한 빛 가림의 영향을 배제하기 위해 생물여과막 반응조를 광생물 반응기 전단에 배치하여 탈질 반응과 유기물 제거 반응이 진행되도록 구성하였다. 이러한 분리배양 시스템을 실제 하수에 적용하여 그 효과를 검증함으로써 미세조류-공배양 시스템의 하수처리 시간을 단축하고 상용화를 위한 대안을 제시하고자 하였다.
바이오매스의 빛 파장별 공배양 광합성 산소 생산 특성을 파악하기 위한 청색, 녹색, 적색 및 무색 LED 조사 연구 결과는 다음과 같다. 1) 공배양 바이오매스에서 빛 투과도는 지수함수적으로 급격하게 감소하지만 광합성에 의한 산소 생산율은 바이오매스 혼합에 의해 대수함수 형태로 완만하게 감소하여 혼합에 의한 효과가 나타났다. 2) 빛 가림과 광 저해 현상에 의한 산소 생산 감소는 청색, 적색 영역에서 녹색 및 무색 영역보다 강하게 나타났다. 특히, 자연광과 유사한 빛 파장의 무색 영역은 광 저해와 빛 가림이 크지 않아 하절기의 강한 빛 조건에서도 광합성 산소 생산에 지장이 없을 것으로 나타났다. 3) 적색 영역의 빛은 광합성 산소 생산속도가 가장 높고 빛 가림에 의한 광합성 저해가 크지 않아 인공광원의 기본 파장으로 이용할 수 있을 것으로 기대된다. 반면, 청색 영역의 빛은 빛 가림과 광 저해 현상이 동시에 강하게 작용하여 광합성 반응이 심각하게 저해되므로 적색광의 보조 광원으로 이용하는 것이 적절한 것으로 판단된다.
실제 하수에서 빛 파장별 하수처리 특성을 파악하였으며, 다음의 연구 결과를 얻을 수 있었다. 1) 영양물질 제거, 산소 공급, 침강성 등 하수처리 효용성 측면에서 적색과 무색 영역의 빛이 녹색과 청색 영역에 비해 양호하게 나타났다. 2) 빛 가림에 의한 광합성 산소 생산 저해를 방지하기 위해서 바이오매스는 500 mg/L 이하로 운전하는 것이 필요하며, 수심 20cm의 광생물 반응기에서 하수 원수를 처리하는 경우 HRT(Hydrolic RetentionTime)는 4일 정도 소요된다. 3) 적색 및 무색 LED에서 질산화-탈질화에 의해 질소가 제거되며(각각 18.8%, 14.1%), 질산화 반응은 미세조류와의 이산화탄소 기질 경쟁에 의해 제약을 받는다.
연속회분식 미세조류-박테리아 공배양 시스템에서 유동담체 충진 효과를 파악하기위한 연구에서는 다음과 같은 결론을 얻을 수 있었다. 1) HRAP 공정에 유동 담체를 충진함으로써 HRT를 4.0일에서 2.5일로 단축하는 효과가 나타났다. 2) 유동 담체 충진으로 질산화 반응이 촉진되며(처리수의 TN의 95% 이상이 NOx-N), 제거된 총질소의 41.4%가 종속영양 탈질 반응에 의해 제거됨으로써 무산소성 유기물 제거 효과가 발생되었다. 3) 광생물 반응기에 유동 담체를 충진함으로써 충진하지 않은 경우보다 질산화 속도가 3.4배, 탈질 속도가 2.0배 향상되어 질소 제거에 효율적으로 적용할 수 있다. 4) 공배양 시스템에서 탈질에 소비되는 BOD는 활성슬러지 공정보다 28.5% ~ 30.3% 절감되어 C/N 비율이 낮은 하수에 유용하게 이용할 수 있을 것으로 기대된다.
미세조류 광합성 산소를 하수처리에 이용하는 공법의 하수처리 시간 단축을 위한 대안으로 종속 및 독립영양 미생물 서식공간을 분리하는 신공정을 개발하였으며 다음과 같은 결론에 도달하였다. 1) 생물여과막(Biofilter : BF) 공정(종속 미생물 서식 공간)과 광합성-질산화(Photosynthesis-Nitrification : PN) 공정(독립영양 미생물 서식 공간)을 결합한 신 공정을 통해 TKN 제거 시간을 약 8배 단축(기존 4일에서 0.5일)할 수 있다. 2) 생물여과막 공정은 NOx-N의 탈질, BOD 산화, 입자 물질 차단 및 가수분해, CO2 공급과 광합성-질산화 공정으로 탁도, BOD 유입을 줄임으로써 광합성-질산화 공정에서 종속 미생물에 의한 빛 가림을 방지하는 역할을 수행한다. 3) 광합성-질산화 공정은 광합성과 질산화 반응이 효과적으로 이루어졌으며(NOx-N이 처리수의 90.0% 이상) 사상성 조류의 우점화로 침강성이 우수하였다. 4) 인 제거율은 TKN 제거율 (95.6%에 비해 낮았은 50.5%로 나타났으며, 생물여과막 공정에서 입자물질 제거(31.2%)와 광합성-질산화 공정의 세포 합성(19.5%)에 의해 제거되었다.
본 연구에서 시도한 유동담체 충진에 의한 질산화율 향상, 종속영양과 독립영양 미생물의 서식 공간 분리를 통한 하수처리 시간의 획기적 단축 기법은 미세조류를 이용한 하수처리 기술의 활발한 보급에 크게 기여할 것으로 기대된다.

Algal-bacterial biotechnologies for municipal wastewater treatment (MWT) can be a novel alternative to intensely save the cost of mechanical aeration of conventional activated sludge treatment (AST) because algae and bacteria are able to interact with their metabolites (photosynthesis O2 and CO2). However, for commercialization of this technology, there are the several technical bottlenecks such as long HRT, shading effect, whether change etc., should overcome. In present study, the various biotic and abiotic parameters to develop new innovative wastewater treatment process with algal-bacterial culture are explored. At first, as a fundamental research, we performed experiments to investigate the effect of three monochromatic LEDs lights (Blue, Green, Red, White wavelengths) on photosynthetic O2 production rate, nutrient removal efficiency, and biomass production of algal-bacterial symbiosis with photobioreactor (PBR) (Chapter 4 and 5). At second, the applicability of moving media to promote biological nitrification reaction of algal-bacterial reactor was evaluated. Thus, the effect of moving media on nutrients removal efficiency and biomass sedimentation as well as HRT shortening was investigated using PBR equipped with blending LED lights of red and white based on the results derived from chapter 4 and 5 (Charter 6). At third, the further experiments were conducted to shorten HRT of algal-bacterial culture for MWT. Then, biofilter reactor (BFR) with short HRT (1~2 hr) was installed in front of PBR to remove the influence of shading effect by heterotrophs as well as to enhance denitrification and organic material removal in MW and PBR was charged with moving media to promote nitrification. Our separation culture system of algal and bacterial applied for real MWT and its benefits such as increase of nitrification and nutrients removal efficiency, HRT shortening, elimination of shading effect, and biomass settle-ability promotion were precisely evaluated in aspects of its commercialization (Chapter 7). Subsequently, the results of chapter 4 were following to: 1) light transmission in water sharply decreased in exponential function and O2 generation rate gradually declined in algebraic function, 2) the influence of shading effect and light inhibition by blue and red wavelengths was stronger than that by green and white wavelengths. Especially, it seem that white wavelength; similar to natural light, will be optimal light source for O2 production under weaken light condition such as winter season because it showed lower influence of shading effect and light inhibition, 3) red light can be utilized as fundamental synthetic light source because it has characteristics of high O2 generation and comparatively low shading effect. However, it might be reasonable that blue wavelength is utilized as auxiliary light due to its high shading effect and light inhibition. The results of chapter 5 in which the effect of above four LED lights on biological parameters of real MWT was explored were following to: 1) higher nutrients removal efficiency, O2 generation and biomass sedimentation rate at red and white light were obtained than those at blue and gree none, 2) it is revealed that the MLSS contents for preventing decrease of O2 generation from shading effect of biomass itself must be maintained at less than 500 mg/L and if HRAP system for real MWT will be operated with the 20-cm depth PBR, it will be take n more than 4 days of HRT, 3)nitrogen removal rates by nitrification/denitrification reaction at red and white PBR were 18.8% and 14.1%, respectively and it may seem that nitrification Is affected by CO2-substrate competition with microalgae. The results of chapter 6 in which the effect of moving media on biological parameters of algal-bacterial symbiosis was explored were following to: 1) nitrification of HRAP process was significantly promoted by moving media resulting in shortening HRT from 4.0 day to 2.5 day (over 95% of effluent TN was NOx-N), 2) nitrification/denitrification reaction by charging moving media was enhanced and more than 41.4% of total nitrogen by nitrification/denitrification reaction was removed and moreover, BOD during denitrification can consumed as electron acceptant resulting in saving the energy to supply O2 for BOD oxidation,3) nitrification and denitrification reaction by charging moving media was promoted as much as 3.4 times and 2.0times, respectively, 4) comparing with AST, 28.5 ~ 30.3% of BOD for denitrificationin PBR was saved and therefore, our PBR can apply for treating the sewage influent with low C/N ratio. Based on above various fundamental and applicable investigations, we challenged to develop a breakthrough algal-bacterial process, which has unique properties of separating inhabitation spaces of heterotrophs and autotrophs in order to dramatically shorten HRT for MWT at chapter 7. The results were following to: 1) our new algal-bacterial process which combined BFR (space of heterotrophs) and PNR for photosynthesis/nitrification (PN, space of autotrophs) shorten the HRT for TKN removal from 4 day to 0.5 day (approximately, 8 times), 2) BFR unit played outstanding roles in NOx-N denitrification and BOD oxidation, block and hydrolysis of suspended solid in sewage, resulting in increase of CO2 supply and decrease of shading effect of heterotrophs to PNR unit, 3) PNR unit also showed high photosynthesis and nitrification rate(over 90% of NOx-N ineffluent TN) and biomass sedimentation was excellent by dominant species of filamentous green algae, 4) phosphorus removal rate(50.5%), 31.2% at BFR and 19.5% at PNR for removing suspended solid and cell synthesis of algae and bacteria, respectively, was low comparing with 95.6% of TKN removal rate. In conclusion, enhancement of nitrification rate by moving bed installation to photosynthesis-nitrification reactor and separation of ecological habitat according to trophic levels are very effective for shortening purification time of wastewater. This new process will be used for facilitating microalgae-bacteria consortium technology.
