These compounds have various phamacological activities such as anti-stress, anti-tumour, anti-inflammatory, anti-aging, and neuro-protective activities. Ginsenosides are classified into two forms; the major form which is highly glycosylated and the minor form which has fewer sugar moieties. The minor ginsenosides are known to have higher activities than other major forms and they are difficult to extract from raw ginseng due to low amounts. Among them, the ginsenoside Rg3 leads attention of scientist because of it anti-metastasis and anti-tumour effects. Therefore, the aim of this study was to produce minor ginsenoside Rg3 from major forms, Rb1 and Rd, by using recombinant Lactococcus lactis expressing heterologous β-glucosidase gene. For this, the nucleotide sequence for β-glucosidase gene (BglBX10) from Flavobacterium johnsoniae UW101 was expressed in L. lactis NZ9000 by using pNZ8008 plasmid (pNZ-BglBX10). When L. lactis NZ9000 harboring pNZ-BglBX10 was cultivated in 100 mL GM17 medium and induced with nisin (1 ng/mL), β-glucosidase activities were detected in cytoplasmic fractions. The enzyme activities were 0.1 units/mL (uninduced) and 0.3 units/mL (induced). The specific β-glucosidase activities in the induced crude extract or purified fraction were 0.002 units/mg or 0.02 units/mg, respectively, showing 10-folds higher purification efficiency. SDS-PAGE analysis of the purified protein by Ni-NTA column gave a single band with a molecular weight of β-glucosidase (87.8 kDa). When the crude cell free extract was reacted with Rb1 and Rd, Rg3 was synthesized after hydrolysis of glucose residues at C-20 position of substrates. When the whole cells of L. lactis harboring pNZ-BglBX10 were reacted with protopanaxadiol type ginsenosides mixture (PPDGM) for 24 h, Rb1 and Rd were converted into Rg3 resulting in 44% of conversion yield. Meanwhile, when the harvested cells were lysated by sonication or permeabilized with 0.5% xylene, the conversion yield increased up to 58 % or 61%, respectively. This study demonstrates that the lysated or permeabilized L. lactis expressing β-glucosidase gene can be used to efficiently produce Rg3 compound in ginseng extract. The cell factory system that developed in this study will provide an alternative method to increase the health functions of Panax ginseng and development of ginseng industry.
