Docking 연구를 동반한 hit 물질인 1a 에 대한 최적화 연구를 통하여 1b와 1d와 같은 강력한 JAK2 억제제들을 발견할 수 있었다. 더욱이 V-shaped kinase 억제제들은 단백질의 활성 부위를 곡선모양으로 채워질 수 있고 pocket 내로 깊게 접근할 수가 있다는 연구를 기반으로 하여 유사한 모양을 갖는 V-shaped 설폰아미드 구조를 갖는 새로운 화합물 7e, 7h 를 개발할 수 있었다. 다음 연구에서는 전립선 암세포의 생존에 필수적인 AR을 타깃으로 선정하였다. 여러 안드로젠수용체 길항제들이 전립선암 치료제로 사용되고 있다. 그러나 현재 사용중인 안드로젠 수용체 길항제들은 거세내성 전립선 암 (CRPC)에는 전혀 효과가 없기 때문에 새로운 구조의 안드로겐 수용체 길항제의 개발이 시급하다. 본 연구에서는 안드로겐 수용체의 활성 부위의 구조를 이용한 structure-based drug design 기술을 통하여 발견한 신규 안드로겐 수용체 길항제인 DIMN의 구조 최적화 연구를 수행하였다. 즉 docking 연구를 통하여 소수성 pocket을 채워줄 수 있는 치환기를 DIMN의 isoquinoline ring에 도입하였으며 clogP 값, 친수성 및 수소결합력의 증가를 위하여 DIMN의 pyridine ring 을 pyrazine, pyrimidine ring으로 바꾸어 다양한 유도체를 합성한 후 생리활성을 측정하였다. 그 중 3d, 3e, 3g 및 6c 물질들이 안드로겐 수용체 길항효과 및 전립선암 세포인 LNCaP가 우수할 뿐 만 아니라 normal cell에 대한 효과에서도 바람직한 결과를 보였다. 그 중 3j는 LNCaP 세포의 성장의 억제에 가장 강한 효과를 보였으며 안드로젠 수용체 길항제로서 우수한 활성을 보였다. Structure-based drug discovery is a complementary approach to high-throughput screening (HTS), facilitating discovery of novel and potential hits from large databases of diverse compounds by docking the compounds into the active site of a target protein. In this study, Janus kinase 2 (JAK2) and androgen receptor (AR) are selected as the target for developing novel anticancer agents. In recent years, genetic, biological, and physiological evidence has established JAK2 inhibitors as effective chemotherapeutic agents for the treatment of many different cancers. For this reason, we sought to identify novel small molecule inhibitors of JAK2. Using Surflex-Dock software, 3,010 compounds were tested in silico for their ability to interact with the JAK2 ATP-binding pocket. Ten highest-scoring compounds were selected and tested for their abilities to inhibit JAK2 activity in vitro. Compound 1a (ethyl 1-(5-((3-methoxyphenyl) carbamoyl)-3-nitropyridin-2-yl) piperidine-4-carboxylate) was identified as the hit of JAK2 inhibitor. Optimization of 1a using docking studies led to the discovery of potent JAK2 inhibitors, 1b and 1d, potent JAK2 inhibitors. Furthermore, as V-shaped kinase inhibitors can curve around the protein backbone and access deep into the pocket, we developed a new series of compounds with a non-linear sulfonamide bond. Compounds 7e and 7h showed a lower IC50 for JAK2 inhibition, validating this design approach. In another research, AR was selected as a target for treating prostate cancer since the significance of AR is vital for survival of prostate cancer cells. Several AR antagonists are clinically prescribed against prostate cancer. Unfortunately, efficiency of existing AR antagonists decrease in a clinical state known castration-resistant prostate cancer (CRPC). To overcome this, a novel AR antagonist candidate called DIMN was discovered by our research group in 2013. With the aim of developing more potent compounds, we designed novel DIMN derivatives based on the docking study and replacing carbon to heteroatom. Encouraging in vitro test results of compounds 3d, 3e, 3g and 6c proved that the new design is successful. Among all the newly synthesized compounds, 3j (IC50 = 0.481 μM) exhibits strongest effect on LNCaP cell growth and also acted as a competitive AR antagonist with selectivity over estrogen receptor (ER). Docking study of 3j fully supported the biological results. Compound 3j is considered to be a novel potent AR antagonist for treating CRPC. The study successfully applied structure-based drug discovery for hit identification, and a docking study for hit optimization. The methods provide a guide for future development of drug candidates through structure-based drug discovery and chemical modifications.
