FFA4 길항제인 AH7614의 투여는 알코올성 지방간에 대한 오메가-3 지방산의 보호효과를 억제하였다. Kupffer 세포에서의 FFA4의 발현을 확인 하였고, 오메가-3 지방산의 투여에 의해 Kupffer 세포에서 TNF-α, COX-2, iNOS, TGF-β, NLRP-3의 발현증가가 감소하였다. FFA4 길항약의 투여는 오메가-3에 의한 활성화된 Kupffer세포의 항염증 효과를 완화시켰다. 이러한 결과는 오메가-3의 지방간 억제효과가 Kupffer 세포의 FFA4를 활성화시킴으로써 항염증 효과를 일으키고, 이를 통해 지방간 억제효과로 매개되었을 것이라 추측할 수 있다. 두 번째로 간세포에서 FFA4의 기능적 역할을 평가하기 위해, 간세포 지방증의 in vitro 모델을 사용했다. Hep3B와 HepG2 간암 세포에서, FFA4 발현이 확인되었고, LXR 효능제인 T0901317이 지질 축적을 유도했으며, 대표적인 오메가-3 지방산인 docosahexaenoic acid (DHA)가 지질 축적을 억제했다. DHA에 의한 억제효과는 FFA4 선택적 길항제인 AH7614 또는 FFA4의 siRNA transfection에 의한 발현억제를 통해 소실되었다. FFA4 효능제인 TUG-891 또한 DHA와 같이 LXR 매개 지질축적을 억제하였다. Abstract Omega-3 polyunsaturated fatty acids (ω-3 PUFAs) have been reported to be beneficial nutrients in various disease states. Recently, the G protein-coupled receptor, FFA4 (also known as GPR120), has been recognized as a sensor for ω-3 PUFAs and its protective function in hepatic steatosis was reported. However, the molecular targets of ω-3 PUFAs have not been fully studied. In this study, the effects of ω-3 PUFAs were investigated by using an alcohol-induced liver steatosis model and a liver X receptor (LXR)-mediated hepatocellular steatosis model. Previously, supplementation of ω-3 PUFAs was reported to be effective for liver fat reduction in acute and chronic alcohol-induced steatosis. Therefore, using an ethanol-induced liver steatosis model, it was investigated whether ω-3 PUFAs protect alcoholic liver steatosis through FFA4 in Kupffer cells. ω-3 PUFAs reduced alcohol-induced increases of triglycerides contents, liver/body weight ratio, and lipid accumulation in hepatocytes and levels of serum AST and ALT. ω-3 PUFAs also reduced alcohol-induced increase of lipogenic SREBP-1c expression and reversed decrease of lipolytic PPAR- expression. Corn oil plenty of saturated fatty acids also showed decrease of lipid accumulation in hepatocytes, but less than ω-3 PUFAs. Administration of AH7614, a FFA4 antagonist, ameliorated ω-3 PUFAs-induced protection on alcoholic hepatic steatosis. FFA4 expression was confirmed in Kupffer cells. ω-3 PUFAs treatment modulated alcohol-induced increase of TNF-, COX-2, iNOS, TGF-, and NLRP-3 in Kupffer cells. Administration of AH7614 ameliorated ω-3 PUFAs-induced anti-inflammatory action on alcoholic activation of Kupffer cells. These results indicate that the anti-inflammatory effects of ω-3 PUFAs may be mediated through FFA4 in Kupffer cells. Second, in order to estimate functional role of FFA4 in hepatocytes, an in vitro model of LXR-mediated hepatocellular steatosis was used. In Hep3B and HepG2 human hepatoma cells, FFA4 expression was confirmed. T0901317, a specific LXR activator, induced lipid accumulation and docosahexaenoic acid (DHA), a representative ω-3 PUFA, inhibited lipid accumulation in Hep3B cells. DHA-induced inhibition was blunted by treatment with AH7614 or siRNA transfection of FFA4. TUG-891, a specific FFA4 agonist, induced inhibition of LXR-mediated lipid accumulation like DHA. siRNA transfection of FFA4 blunted the TUG-891-mediated inhibition also. SREBP-1c, a key transcription factor for lipogenesis, was induced by T0901317 treatment. The SREBP-1c induction was also inhibited by DHA or TUG-891 at mRNA and protein levels. DHA-induced suppression of SREBP-1c expression was again blunted by FFA4-knockdown in the hepatocytes. DHA suppression of LXR-induced lipid accumulation was also observed in human HepG2 cells and primary hepatocytes from mice. The suppression was blunted by AH7614 treatment in Hep3B and HepG2 cells and was not observed in primary hepatocytes from FFA4 deficient mice. These results indicate that ω-3 PUFAs function to protect LXR-induced steatosis through FFA4 in hepatocytes. Therefore, the present study suggest that ω-3 PUFAs function to protect alcoholic hepatic steatosis by activation of FFA4 in Kupffer cells and to protect non-alcoholic hepatic steatosis by activation of FFA4 in hepatocytes.
