As the amount of plastic use increase, the amount of discarded plastic in aquatic ecosystems also increase. These abandoned plastics decompose to microplastics(MPs), which size are less than 1mm, and consequently, these materials expose to aquatic organisms. Furthermore, microplastics can unite with other water pollutants and influence the health of the organisms. In this study, we evaluated the combined toxicity of nickel and microplastics on the cladoceran Daphnia magna. We used two sorts of a microsize polystyrene sphere (one had a carboxyl functional group, and the other had no functional group). Toxicity tests were conducted: 1) toxicity tests of single chemical (nickel and microplastics), 2) variable Ni-fixed MPs 5mg/L, 3) fixed Ni 3mg/L-variable MPs, 4) variable Ni-variable MPs. Endpoint of acute toxicity test was immobilization of D. magna. The values of EC50 were calculated from the results and, the values of EC50mix were calculated using Toxic Unit(TU) model for mixture toxicity tests. As a result, the value of EC50 of Ni was 3.85mg/L and the values of EC50 of PS and PS-COOH were 42.78 and 36.65mg/L, respectively. When MPs had funtional group, toxicity of MPs was increased. For mixture toxicity tests, the values of EC50mix of variable Ni-fixed PS and PS-COOH 5mg/L were 1.33 and 0.96, respectively. The values of EC50mix of fixed Ni 3mg/L-variable PS and PS-COOH were 1.16 and 1.05, respectively. The values of EC50mix of variable Ni-variable PS and PS-COOH were 1.07 and 0.78, respectively. We found that the combined toxicity of nickel with the MPs was greater than that of nickel or MPs alone. We also compared the two complex toxicity with functional groups and confirmed that the combination of nickel and MPs bound to a carboxyl group was more toxic to D. manga than the combination of nickel and MPs without a functional group. Meanwhile, nickel concentrations in D. magna increased in the presence of MPs, especially PS-COOH, compared when MPs didn’t exist. This is probably due to increase of nickel uptake for the absorption of nickel into the MPs. We also conclude this was likely due to positive nickel ions binding to the negatively charged carboxyl groups, which eventually enhanced the uptake of nickel by the cladoceran. The results imply that the toxicity of aquatic pollutants can be increased by the presence of microplastics. Therefore, we suggest that this phenomenon may occur with other pollutants and microplastics, and further research is needed to investigate the complex toxicities of other pollutants and microplastics.
