Interestingly, both proteins act as scaffold proteins by connecting the Ras to Raf in mitogen-activated protein kinase (MAPK) signaling pathway. These results suggest that DUF1 functions as a negative regulator of MAPK signaling by interfering with the scaffolding functions of PHB1 and SAMHD1, and consequently controlling host cell growth and survival.
