미생물을 활용한 전통적인 발효과정에서 생성되는 생리활성 화합물 (Biologically active compounds)은 미생물에서 동•식물에 이르기까지 다양한 종에서 발견된다. 이러한 생리활성 화합물을 이용한 종자 배양물의 사용은 발효 과정에서 생산물의 안정성을 향상시키기 위한 중요한 전구체로 간주되고 있다. 이는 항균 펩타이드 및 미생물 효소를 생산할 수 있는 박테리아는 천연 자원으로써 김치와 같은 전통 발표 식품이 질병 치료 및 바이오 산업에 응용에 이용될 수 있음을 시사한다. 특히, bacillus 균주는 다루기 쉽고 성장속도가 빠르며, 배지에서 대량의 항균 펩타이드와 미생물 효소를 생산할 수 있기 때문에 생리활성 화합물 연구에 있어서 매우 흥미로운 미생물로 알려져 있다. 본 연구는Bacillus 균주의 생리활성 화합물을 분석하고 이들의 바이오 산업에서의 응용 가능성을 평가하였다. 연구에서 사용 된 5가지의 박테리아 균주는 200 가지 이상의 박테리아 균주 라이브러리에서 스크리닝 되었으며, 세균 계통 분리학의 Bergey 's Manual을 참고하여 확인하였다. Bacillus 균주의 형태학적 특성은 16S rRNA 유전자 서열 분석에 의해 확인하였다. 균주가 생산하는 생리활성 화합물의 정제는 diammonium sulfate 분획, 투석, 한외여과, molecular weight cut off (MWCO) 그리고 순차적 크로마토그래피 분리 기술들을 사용하였다. 항균 펩타이드 (AMPs)는 잠재적인 신규 치료제로서, 광범위한 항균 활성 및 강력한 항생제 특성을 갖는 저분자량 단백질이다. 약물 내성의 급격한 증가는 항생제 치료를 어렵게 하는 요인으로, 내성의 증가에 따라 전통적인 항생제를 대체 할 수 있는 새로운 치료제의 개발 필요성이 요구되고 있다. 본 연구에서, 새로운 치료제의 개발을 위한 생리활성 화합물들을 생산하기 위해 다양한 항균 펩타이드들을 분석하였다. 항균 펩타이드는 Bacillus amyloliquefaciens K14 (CSpK14) 및 Bacillus subtilis CSB138 (p138c)에서 생산되었다. 생화학적 특성 분석 결과는 다음과 같다. CSpK14는 약 4.6 kDa의 분자량을 가지며 pH 6.0-11.5 및 온도 80 °C까지 안정성을 가졌다. p138c는 약3 kDa의 분자량을 가지며 50 °C에서 내열성을 보였고 pH 5.8에서 11까지 안정하였다. CSpK14 (His-Tyr-Asp-Pro-Gly-Asp-Asp-Ser-Gly-Asn-Thr-Gly)의 아미노산 서열은 alpha-2-macroglobulin 및 ligand-gated channel 단백질과 일정부분 동일성을 나타내었다. VRSA 균주를 대상으로 항균활성 실험을 한 결과, CSpK14 / oxacillin 과 CSpK14 / ampicillin 의 최소 억제 농도 (MIC)는 각각 8-64배, 2-16배까지 감소하였다. CSpK14 / oxacillin (24 시간에서 2.29–2.37 Δlog10CFU/mL)과 CSpK14 / ampicillin (24 시간에서 2.30-2.38 Δlog10CFU/mL)의 살인 동역학은 2배 이상 증가하였고 FIC 지수는 0.05이하의 시너지 효과를 나타내었다. 반코마이신 내성 포도상 구균 (VRSA) 균주 및 반코마이신 내성 장구균 (VRE)에 의해 형성된 바이오 필름은 이 시너지 효과에 의해 완전히 감소되었다. 항균 펩티드 p138c 생산은 후기 지수 성장 단계에서 최대화되었다. 실험결과 19.15 fold와 3.2%의 회수율을 보이며 4800 arbitrary unit 달성이 가능했다. p138c는 Gly-Leu-Glu-Glu-Thr-Val-Tyr-Ile-Tyr-Gly-Ala-Asn-Met-X-Ser의 N-말단 아미노산 서열이 확인되었다. VRSA에 대한 p138의 효능은 p138c가 옥사실린, 암피실린 및 페니실린 G와 함께 작용하였을 때, 각각 4배, 8배, 16배의 향상 효과를 보였다. 옥사실린, 암피실린 및 페리실린 G가 처리된 p138c의 시너지 테스트 (FIC 지수)는 각각 0.3125, 0.25 및 0.09 였으며, 각각 향상된 항균활성을 나타내었다. 따라서, 이러한 항균 펩타이드 (AMPs)는 항생제 내성을 극복하고 MDR 감염 치료에 적합한 작용물질로 복합적인 치료 개발에 도움이 될 수 있다고 판단 할 수 있다. 미생물인 mannanases는 식물 조직의 복합적 다당류를 manno-oligosaccharides 와 mannose 와 같은 단순한 분자로 가수분해하기 때문에 생체공학적으로 중요하다. Mannanases 는 펄프, 종이, 식품 및 의약품 등과 같은 바이오 산업 분야에서 관심이 부각되고 있는 효소이다. 따라서, 본 연구에서는 로커스트콩껌에서 생산된 미생물인 mannanases을 연구하고자 하였다. Maananases는 Bacillus sp. CSB39 (MnCSB39) 와 Bacillus subtilis subsp. inaquosorum CSB31 (MnB31) 로 부터 전기영동 균질을 통해 정제되었다. 총 571.14 U/mL의 MnCSB39가 생성되어 25.47% 수율과 19.32배의 순도를 나타내었다. MnCSB39는 약 30 kDa의 분자량으로 Bacillus mannanases 중에서 가장 적은 분자량을 가졌다. MnCSB39는 pH 7.5와 70 ℃에서 최적의 활성을 보였으며, 5.0–15% (w/v) NaCl 조건에서 55% 이상의 활성을, 37 ℃에서 60 분 동안 배양 한 후 초기 활성도는 93% 이상을 나타내었다. MnCBS39 활성은 trypsin 및 proteinase K 처리에 의해 영향을 받지 않았으며, 최대 3M urea에서 80% 이상의 활성을 나타내었다. 로커스트콩껌을 가수분해하기 위한 활성 에너지는 26.85 kJmol−1 이었고 Kcat 은 142.58 × 104 s−1이었다. Km 과 Vmax는 각각 0.082 mg/mL 과 1099 ± 1.0 Umg−1 이었다. ΔH, ΔG, ΔS, Q10, ΔGE-S, 그리고 ΔGE-T 는 생성물의 자발적인 형성과 효소 반응성의 높은 가수분해 가능성을 확인하였다. 가수분해 실험 (TLC) 결과, mannobiose가 주된 최종 생성물이라는 것을 확인하였는데, 추가적인 미생물 mannanase 연구를 통해 로커스트콩껌에서 생성된 알칼리성 mannanase (MnB31)가 21.51% 수율과 1796.13 Umg-1의 특정 활성을 나타내면서17.92배로 정제됨을 확인하였다. 생화학적 연구 결과에서는 MnB31가 pH 12.5 의 KCl/NaOH 완충액에서 최대 효소 활성을 나타내었으며, pH 5.8-12.5에서 안정적이며 60 oC에서 최적 활성을 보이고 65 oC에서 열 안정성을 보이는 것을 확인하였다. SDS-PAGE 전기영동 결과, 분자량은 약 47kDa으로 확인되었다. MnB31의 N-말단 아미노산 서열은 Ala-Leu-Tyr-Glu-Thr-Ile-Phe-Ala-Leu-X 로 Co2+, Mn2+, Na+ 및 K+ 에 의해 최대 활성화되었다. Km 과 Vmax는 각각 0.043 mgmL-1 과1046 ± 3.605 Umg-1 이었다. LBG의 가수분해를 위한 활성화 에너지 (Ea)는 31.36 kJmol-1 이었고 Kcat 값은 156.9×104 s-1이었다. LBG 가수분해를 위한 ∆H (28.59 kJmol-1), ∆G (42.38 kJmol-1), ∆S (-41.39 Jmol-1K-1), Q10 (1.40), ∆GE-S (-8.697 kJmol-1) 및 ∆GE-T (-48.22 kJmol-1)와 같은 열역학 인자는 자발적 과정으로 발생되었다. MnB31은 endo-acting의 특성을 가진다. MnCSB39와 MnB31은 광범위한 pH와 온도 범위, 다중 스트레스 내성을 가지고 있으며, 열역학적으로 가능한 반응을 촉매 하기 때문에 바이오 산업 촉매제로 개발 될 가치가 있다고 생각된다. Endoglucanase는 식물 바이오매스 분해에 필수적인 효소이다. 그러나 대부분의 응용과정에서 실질적으로 높은 비용과 낮은 촉매율이 Endoglucanase의 단점으로 지목되고 있다. 본 연구를 통해, β-glucanase의 촉매작용 및 화학적 상호작용을 유도하여 높은 특수성을 설명하였다. 본 연구에 사용된Glucanases는 Bacillus sp. CSB55 (GluB55)와 Bacillis subtilis CSB31 (GluCB31)에서 생성되었으며, 정제된 GluB55 효소는 약40kDa의 분자량과 pH 7.5와 55oC에서 최대활성을 보여주었다. 또한, GluB55는 carboxymethyl cellulose (CMC)를 가수 분해 할 수 있으며 chitin에 대해 50 % 이상의 활성도를 나타내었다. GluB55는 pH 5.8 ~ 11.0에서 70% 이상의 잔류 활성을 보유하고 있었고, GluB55 는 60 oC 에서 100%의 내열성을 보였으며 70 oC 에서는 68%의 내열 효율을 보였다. 효소활성은 Co2+ (158.6%), Zn2+ (211.1%), Mn2+ (264.4%) 및 Ba2+ (211.4%)에 의해 증가 되었다. GluB55는 EDTA에서 85.4 %의 활성을 보였으며 EGTA에서는 83.7 %의 활성을 보였다. Edman 분해 방법을 이용하여 16개의 N-말단 아미노산 서열; A-N-P-E-L-V-N-X-Q-A-X-X-A-X-Q-G을 확인하였다. 운동 연구는 0.022 mg/mL 의 Km 및 Vmax of 994.56 ± 3.72 U/mg 의 Vmax를 나타냈다. 또 다른 실험에서, GluCB31은 8.83%의 수율 및1066.37 U/mg의 특성 활성으로 17.68배까지 정제되었다. GluCB31의 생화학적 특징을 확인한 결과, GluCB31은 35kDa의 분자량을 가지며 pH 7.5 와 50 oC에서 최적 활성을 가지는 것을 확인하였다. GluCB31은 광범위한 pH (6.5-10.0)와 온도 허용 오차를 보여 주었다. 고정화된 효소는 광범위한 pH범위 (4.6-10.0)와 최대 60 oC 까지의 온도범위를 나타내었다. Line weaver-Burk Plot에 의한 Kinetic 연구를 통해, GluCB31가 1293.33 ± 2.51 U/mg의 Vmax 와 0.0183 mg/mL의 Km을 나타내는 것을 확인하였다. 효소활성은 Tween-20 (106.7 %), Tween-80 (111.6 %), Triton X-100 (142.3 %), SDS (135.5 %), Mg++ (185.7 %), Cu++ (167.6 %), Zn++ (153.7 %), Mn++ (106.3 %), Ba++ (181.9 %), Ni++ (107.2 5)에 의해 활성화되었고, 반면에 Fe++ (15.8 5), β-mercaptoethanol (46.8 %), EDTA (54.5 %)에 의해 억제효과를 나타내었다. GluCB31의 N-말단 아미노산 서열은F-P-G-V-M-A-L-V-M-A-I-X-A로 확인되었다. 따라서, GluB55와 GluCB31의 이러한 특성들은 촉매 활성의 다양성을 보여주고 있으며, 생명 공학의 다양한 측면에 적용하여 바이오 산업의 효소로써 개발 될 가치가 있다는 것을 보여주고 있다. 결론적으로, 항균 펩타이드는 다약제 내성 감염을 극복하기 위한 복합 약물 치료제의 주요 후보로서 바이오 산업에 응용될 가치가 있다. 미생물유래 효소에 대한 이해는 다양한 효소 단백질의 촉매 특이성이 바이오 산업 전반에 걸쳐 영향력을 가지고 있으며, 이것들이 식물 바이오 매스 분해에 어떻게 기여하는지에 대한 다양한 방법 등을 제공한다.

Biologically active compounds, relying on traditional fermentation of natural microbial flora and natural conditions, have been found in different living organisms ranging from microorganisms to animals and plants. The use of seed culture consisting of such agents is regarded to be an important precursor to improve the safety and stability of the products in fermentation process. This emphasize the potential use of traditional fermented foods such as kimchi (a popular Korean food) as natural resources of bacteria, which is capable of producing antimicrobial peptides and microbial enzymes for therapeutic and bio-industrial applications. Bacillus is an interesting microorganism to be investigated as a potential biologically active compounds producer because it is easy to handle (safety), fast grower and ability to produce large amount of antimicrobial peptides and microbial enzymes in the production media. This dissertation was designated to evaluate the biologically active compounds induced by Bacillus strains and applications thereof in the therapeutic and bio-industrial sector. Five bacterial isolates used in our study were screened from more than 200 bacterial strains library and identified according to Bergey’s Manual of Systematic Bacteriology. Morphological properties suggesting the parent strains that belonged to Bacillus were further confirmed by the 16S rRNA gene sequence analysis. Purification strategy used in this study was diammonium sulfate fractionation, dialysis, ultrafiltration, molecular weight cut off (MWCO), and sequential chromatographic separation, which were slightly modified in each experiment. Antimicrobial peptides (AMPs) are low molecular weight proteins with broad spectrum antimicrobial activity and potent antibiotic properties that demonstrate as potential novel therapeutic agents. Rapid increase in drug-resistant infections is a serious challenge to antimicrobial therapies, therefore the development of alternative to conventional antibiotics has alarmed. In this study the potent AMPs were procured to make them especially interesting compounds for the development of novel therapeutics. Antimicrobial peptides were produced from Bacillus amyloliquefaciens K14 (CSpK14) and Bacillus subtilis CSB138 (p138c). Biochemical characterization was performed and the results were as follows; CSpK14 had molar mass of ~4.6 kDa and stable at pH 6.0–11.5 and temperature up to 80 °C. p138c was ~3 kDa and thermo-tolerant as high as 50 °C and stable at pH 5.8 to 11. The amino acid sequence of CSpK14 (His-Tyr-Asp-Pro-Gly-Asp-Asp-Ser-Gly-Asn-Thr-Gly) shows some degrees of identity with alpha-2-macroglobulin and ligand-gated channel protein. The minimum inhibitory concentrations (MICs) of CSpK14/oxacillin and CSpK14/ampicillin were reduced by 8- to 64-fold and 2- to 16-fold, respectively. The killing kinetics of CSpK14/oxacillin (2.29–2.37 Δlog10CFU/mL at 24 h) and CSpK14/ampicillin (2.30–2.38 Δlog10CFU/mL at 24 h) was >2-fold and fractional inhibitory concentration (FIC) index ˂0.5 revealed synergy. The biofilms formed by vancomycin-resistant Staphylococcus aureus (VRSA) strains and vancomycin-resistant Enterococci (VRE) were reduced completely by this synergistic combination. Antimicrobial peptide p138c production was maximized in the late exponential growth phase. A total of 4800 arbitrary unit (AU) with 19.15-fold and 3.2 % recovery was possible at the end of purification. p138c had N-terminal amino acid sequence of Gly-Leu-Glu-Glu-Thr-Val-Tyr-Ile-Tyr-Gly-Ala-Asn-Met-X-Ser. Potency of p138c against VRSA showed a 4-, 8-, and 16-fold improvement when p138c was synergized with oxacillin, ampicillin, and penicillin G, respectively. Synergy testing (FIC index) of p138c with oxacillin, ampicillin, and penicillin G was 0.3125, 0.25, and 0.09, respectively revealed enhanced kill kinetics. Therefore, such agents (AMPs) could be beneficial for the development of a combination therapy, acknowledged as the prime approach to overcome antibiotic resistance and to develop a suitable agent for the treatment of MDR infections. Mannan-degrading enzymes are attracting growing interest in bio-industrial applications, such as the pulp and paper, food, and pharmaceutical industries. Here, we studied the microbial mannanases produced in locust bean gum and purified to electrophoretic homogeneity from Bacillus sp. CSB39 (MnCSB39) and Bacillus subtilis subsp. inaquosorum CSB31 (MnB31). A total of 571.14 U/mL of MnCSB39 was produced that resulted in 25.47 % yield and 19.32-fold purity. MnCSB39 had a molecular mass of approx. 30 kDa which is the lowest among the Bacillus mannanases. MnCSB39 had optimal activity at pH 7.5 and temperature of 70 °C. MnCSB39 showed ˃55 % activity at 5.0–15 % (w/v) NaCl, and ˃93 % of the initial activity after incubation at 37 °C for 60 min. MnCBS39 activity was not affected by trypsin and proteinase K treatment. It demonstrated ˃80 % activity in up to 3 M urea. Activation energy for locust bean gum hydrolysis was 26.85 kJmol−1 with a Kcat of 142.58 × 104 s−1. It had Km and Vmax of 0.082 mg/mL and 1099 ± 1.0 Umg−1, respectively. ΔH, ΔG, ΔS, Q10, ΔGE-S, and ΔGE-T supported the spontaneous formation of products and the high hydrolytic efficiency and feasibility of the enzymatic reaction. Hydrolysis experiment (TLC) showed mannobiose as the principal end-product. In a separate microbial mannanase study, an extremely alkaline mannanase (MnB31) produced in locust bean gum was purified to 17.92-fold with a 21.51 % yield and specific activity of 1796.13 Umg-1. Biochemical study demonstrated maximum enzyme activity with KCl/NaOH buffer at pH 12.5 and stable at pH 5.8-12.5 and optimally active at 60 oC and thermostable to 65 oC. SDS-PAGE showed a molecular mass of ~47 kDa. The N-terminal amino acid sequence of MnB31 was Ala-Leu-Tyr-Glu-Thr-Ile-Phe-Ala-Leu-X. MnB31 was highly activated by Co2+, Mn2+, Na+, and K+. It had a Km and Vmax of 0.043 mgmL-1 and 1046 ± 3.605 Umg-1 respectively. The activation energy (Ea) for LBG hydrolysis was 31.36 kJmol-1 with a Kcat value of 156.9 × 104 s-1. Thermodynamic parameters such as, ∆H (28.59 kJmol-1), ∆G (42.38 kJmol-1), ∆S (-41.39 Jmol-1K-1), Q10 (1.40), ∆GE-S (-8.697 kJmol-1), and ∆GE-T (-48.22 kJmol-1) for LBG hydrolysis suggests this as a spontaneous process. MnB31 had endo-acting nature. MnCSB39 and MnB31 had wide pH and temperature ranges, multi-stress tolerant and catalyze a thermodynamically possible reaction, thus deserved to be developed as bio-industrial catalyst. In this study, a plausible explanation is thus proposed by elucidating the catalytic behavior, chemical interaction leading to the high specificity of β-glucanase. Microbial glucanases were produced from Bacillus sp. CSB55 (GluB55) and Bacillis subtilis CSB31 (GluCB31). Biochemical characterization of the purified GluB55 enzyme revealed the molecular mass of approximately 40 kDa and the optimum activity at pH 7.5 and 55 oC. GluB55 can highly hydrolyze carboxymethyl cellulose (CMC) and showed >50 % activity on chitin. GluB55 retained more than 70 % residual activity at pH 5.8 to 11.0. GluB55 carried its 100 % thermo-stability as high as 60 oC and demonstrated about 68 % residual activity at 70 oC. The enzyme activity was stimulated by Co2+ (158.6%), Zn2+ (211.1%), Mn2+ (264.4%), and Ba2+ (211.4%). GluB55 showed 85.4 % activity on EDTA and 83.7 % activity on EGTA. Edman degradation method demonstrated 16 N-terminal amino acids sequence namely; A-N-P-E-L-V-N-X-Q-A-X-X-A-X-Q-G. Kinetic study showed Km of 0.022 mg/mL and Vmax of 994.56 ± 3.72 U/mg. In another experiment, GluCB31 was purified to 17.68-fold with an 8.33 % yield and specific activity of 1066.37 U/mg. Biochemical properties of GluCB31 were performed and the results are as follows; molecular mass of 35 kDa with an optimum pH at 7.5 and temperature at 50 oC. GluCB31 showed wide range of pH (6.5-10.0) and temperature tolerability. Immobilized enzyme possessed broad pH range (4.6-10.0) and temperature up to 60 oC. Kinetic studies showed the Vmax of 1293.33 ± 2.51 U/mg and Km of 0.0183 mg/mL when determined by Line weaver-Burk Plot.
