Enzyme-assisted extraction 법의 경우에도 추출물의 항균성, 항산화성 등의다양한 생화학적 지표가 고전적 추출법의 결과와 비교하여 개선된 점을 관찰할 수 있었다. 한편 Agaricus bisporus 및 Flammulina velutipes의 enzyme-assisted extraction 법의 경우, 고전적 추출법과 비교하여 추출물의 HeLa 세포주에 대한 항암성이 90-370 배 증가하였으며, 해당 추출물의 일반세포에 대한 독성은 매우 낮다는 점을 확인하였다. 세부 연구 결과, 조추출물 중 항암성 성분은 다당류로 예측되었으며, 조추출물의 HeLa 세포주에 대한 IC50은 96 μg mL-1 및 34 μg mL-1로 측정되었다. 해당 다당류에 대한 세부적 분리가 진행 중이며, 한편 조추출물의 다양한 성분 간의 상승 작용과 같은 효과 역시 검토 중이다. 종합적으로 볼 때, 본 연구에서 개발된 추출법은 기존 방법과 비교하여 경제성등의 다양한 측면에서의 개선을 이루었으며, 또한 항암성의 대폭 증대와 같은 저가치 버섯의 부가가치 상승방안으로 활용될 수 있을 것으로 사료된다.

Edible mushrooms were recognized as functional foods and as a source for the development of medicines and nutraceuticals. With the increasing awareness on the health benefits of mushrooms, production of mushrooms is continuously increasing over time. The global mushroom production has increased with an annual output of 2.18 to 3.14 million tons over a period of ten years (1997-2007). The demands are growing on the fact that mushroom encompasses a huge diversity of biomolecules not only nutritional but also their medicinal properties. However, most of the previous studies mainly focused on the research of wild and medical mushrooms, which a have high content of bioactive compounds, diverse therapeutical effects, but a low production. Despite the relatively less amount of nutrients, cultivated mushrooms have produced in a large quantity, and stable quality through the year. So far, cultivated mushroom did not receive much attention from researchers. Therefore, it is imperative to promote the use of cultivated mushrooms as a source of nutrients and nutraceuticals. This study is to develop an efficient, advanced extraction method on enhancing bioactive compounds and their bioactivities from the predominant cultivated mushrooms in Korea. The systematic optimization of the probe-type ultrasonication, bath-type sonication and enzyme-assisted extraction methods have been developed, and a comparison of their effectiveness versus conventional approaches have been demonstrated. The bioactive components in the extracts from different extraction methods have been characterized using biochemical characterization, HPLC, as well as GC-MS analysis. It was found that the probe-type ultrasonication extraction on cultivated mushrooms could efficiently reduce the extraction time from 24 h to 2 min and the content of total phenolics, flavonoids, and antibacterial activities improved remarkably relative to conventional extraction. On the basis of the optimized condition, probe sonication method was also applied to commercial mushroom capsules to test their bioactive properties and antimicrobial activities. The results indicated that the bioactive properties of the mushroom powders, when used as marketed capsules, was very less. The ultrasonication processed mushroom samples showed a significant enhancement of total phenolics, flavonoids, and antioxidant activity. The micro size reduction of the mushroom powders leads to significant enhancement of antiviral properties compared to antibacterial and antifungal properties. Similar trends were also observed in the enzyme-assisted extraction method, which gave a high concentration of bioactive components, stronger antioxidant, and antimicrobial activities compare with those from conventional methods. The enzyme assisted extracts of Agaricus bisporus and Flammulina velutipes exhibit significantly enhanced anticancer activity against HeLa cells with no side effects (90-370 folds enhancement compare to conventional extraction). A further isolation, purification, and identification of macromolecules with anticancer activity from F. velutipes have been conducted. The results found that the crude enzyme extracts and the crude polysaccharides from F. velutipes could potently inhibit the proliferation of cancer cell lines in a dose-dependent manner, with IC50 of 96 μg mL-1 and 34 μg mL-1, respectively. Nonetheless, no apparent antiproliferation activity was observed in the two purified polysaccharide fractions, which indicated that the antiproliferation activity of the crude polysaccharides might be due to the synergistic effect of two polysaccharide fractions or the other unknown compounds, like proteins, phenolics. Overall, these findings provide a new technology for large-scale industrial processing of mushroom for superior human benefits.
