Therefore, discovery of new antibiotics including antimicrobial peptides (AMPs) and flavonoids are emerged. Antimicrobial peptides (AMPs) are small peptides that part of the innate immunity of many higher organisms. They also possess immunomodulatory effects as well as high antimicrobial activity. There has been growing interest in AMPs, which can fight against drug-resistant bacteria. We designed 12-mer potent AMP from the N-terminal sequence of insect AMP, papiliocin. Among nine peptides, Pap12RW-7 showed high potency against various Gram-negative pathogenic bacteria and showed potent anti-inflammatory activities in LPS stimulated RAW cell and its mechanism involve the toll-like receptor 4 pathway. Furthermore, both in serum and lung lysate, Pap12RW-7 peptide inhibited the amount of inflammatory cytokines TNF-α and IL-β in E.coli induced mouse septic shock model. Pap12RW-peptides was able to significantly reduce the levels of bacterial count in lung, kidney and liver lysate of mouse model. Flavonoids are the most abundant polyphenolic compounds present in the human diet in vegetables, cereals, and fruits. Finding novel types of anti-tuberculosis(TB) therapeutics are also necessary due to an increase in the drug-resistant Mycobacteria tuberculosis(MTB). Here, we found that 3,6-DHF(dihydroxyflavone), a chemically modified form of flavone basic scaffold, have anti-mycobacterial effects on H37Rv, MDR, and XDR clinical isolates. Since MTB cause lung inflammation that induces TB pathogenesis, anti-inflammatory effects of 3,6-DHF in MRC-5 cells and dendritic cells stimulated by interferon-γ and lipopolysaccharide (LPS) respectively were investigated. The mRNA levels and quantity of inflammatory cytokines were inhibited by 3,6-DHF, and inflammation mediated MAPKs were suppressed. Therefore, AMPs and flavonoid 3,6-DHF can be a useful therapeutic drug candidate.
