SAMs이 가지고 있는 특징의 장점은, i) 구조적인 특징이 매우 잘 정립되어 있으며, ii) 다양한 기능기의 표면밀도를 쉽게 조절가능하며, iii) 선택적인 반응물질 이외의 물질의 접근을 막아주며, iv) 다양한 분석 장비에 의한 분석이 가능하다는 점이다. 또한, SAMs을 형성하고 있는 티올분자를 Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry(MALDI TOF-MS)를 통하여 분석 SAMDI-TOF MS를 이용해 티올 분자의 분자량 변화를 확인함으로써 단분자층 위에서 발생하는 화학/생화학적 변화를 빠르고 직접적으로 분석이 가능하다. 이러한 다양한 장점을 가지는 SAMs의 특징을 보다 빠르고 손쉽게 분석하기 위하여, 첫 번째 주제는 MALDI-TOF MS를 사용하여 SAMs을 형성하고 있는 티올분자간의 자가교환 속도를 측정하기 위한 새로운 방법을 개발 및 확인하였다. 티올분자간의 자가교환을 측정하기 위하여, 분자 구조적 특징은 같지만 중수소치환반응을 통하여 분자량이 다른 티올분자를 합성하였다. 금 표면에 중수소 치환된 티올분자로 SAMs 형성 후 중수소 치환을 하지 않은 티올 분자가 들어있는 용액에 넣어 티올분자간의 자가교환을 SAMDI-TOF MS를 이용하여 확인하였다. 연구의 조건은 i)처리한 시간, ii)고정온도, iii)다양한 농도, iv)용매조건, 그리고 v)구조적 차이 등 다양한 조건에서 확인하였다. 이러한 실험적 방법을 검증하기 위하여, molecular dynamics simulations을 통하여 자가교환의 분자적 기전을 확인하였다. 연구결과, THF 용매 조건에서 단분자층의 안정성에 큰 영향을 주어 티올분자간 자가교환의 초기속도가 매우 빠르게 나타났다. 이러한 결과에 착안하여, 금 표면에 형성된 자가조립단분자층의 티올 분자의 탈착을 촉진하여 matrix 분자와 티올 분자간의 co-crystallization을 유도하여 다양한 기능기 및 바이오물질을 티올분자에 고정화 시켜 THF를 이용한 SAMDI의 효율 증가를 확인하였다. 새롭게 개발된 방법은 SAMDI의 효율을 향상시킴으로써 다양한 분야에 사용이 될 수 있을 것이다. 두 번째 주제는 다양한 저분자 물질을 Converted Graphene-like Monolayers(CGM) 기판을 사용하여 matrix없이 LDI-TOF MS로 직접적으로 분석할 수 있는 방법을 연구하였다. CGM을 형성하기 위하여 biphenyl-4-thiol을 이용하여 금 표면 위에 SAMs 형성 후 e-beam과 고온열처리 과정을 진행하여 CGM을 형성한다. 형성된 CGM의 분자 구조적 특징을 라만분광기로 측정하여 확인하였다. CGM 기판을 이용하여 다양한 저분자 물질을 LDI-TOF MS로 분석 및 확인했으며, 다른 graphene 물질과 실제 측정 후 비교분석 결과, i) 더 우수한 signal to noise, ii) 최저검출농도, iii) 낮은 백그라운드 신호를 확인하였고, iv) 재사용 가능성을 증명 하였다. 또한, CGM 기판 위에서 cytochrome P450 2A6 효소를 이용하여 coumarin의 효소반응을 다른 정제단계 없이 직접적으로 표면에서 분석이 가능 확인하였다. 세 번째 주제는 여러 개의 miRNAs를 빠르게 고감도 다중검지 및 정량적 분석을 위하여 중수소 치환된 신호 리포터의 신호증폭 기술을 사용하여 연구하였다. 이 방법은 miRNAs를 검출할 수 있는 DNA와 많은 양의 신호 리포터로 코팅된 금 나노입자를 사용하여 진행하였다. 이러한 신호 리포터들을 amplification tags (Am-tags)라고 명명하였다. LDI-TOF MS로 분석 시 하나의 표적 miRNAs로 인해 나노입자 위의 많은 양의 신호 리포터가 검출되는 신호증폭이 현상이 발생한다. Am-tags의 경우 중수소 치환된 개수에 따라 0, 4, 8, 12로 구분하였으며, 각 각의 Am-tags를 사용하여 서로 다른 4가지의 miRNAs 표적을 빠르게 고감도 다중검지를 할 수 있었다. 또한, 각 각의 중수소 치환된 Am-tags의 신호강도를 비교함으로써, 표적 miRNAs농도의 상대적인 정량을 확인 할 수 있었다. 개발한 방법의 검증을 위하여 혈관세포(HUVECs)에 CuSO4를 처리하여 변화되는 다양한 표적 miRNAs의 발현 양을 다중검지 및 정량 연구를 진행, 각 변화된 miRNAs를 개발된 연구방법과 qRT-PCR방법으로 검출된 양을 비교결과, 유사한 결과를 확인함으로써 개발한 방법이 고감도 다중검지 및 정량이 가능한 방법임을 검증하였다.

In this thesis, I describe new analytical tools for self-assembled monolayers (SAMs) on gold and their applications. The SAMs are widely used in various research fields in surface sciences due to their various advantages over other surface platforms: i) well-known structural properties, ii) ease control of surface-functional group density, iii) flexible immobilization strategy of various (bio)molecules, and iv) compatibility to various analytical measurements. Especially, combination of SAMs of alkanethiolates on gold with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI TOF-MS), termed SAMDI-TOF MS, permits identification of the mass changes on alkanethiolates, and thus, rapid tracking of chemical/biochemical conversions on the monolayer. Firstly, I investigated the self-exchange rates of alkanethiolates in SAMs on gold using MALDI-TOF MS in chapter I. In particular, I prepared a deuterated alkanethiol that has the same molecular properties as the non-deuterated alkanethiol but a different molecular weight. SAMs consisting of deuterated alkanethiolates were immersed in a solution of the non-deuterated alkanethiol, and the influences of the immersion time, temperature, concentration, and solvent on the self-exchange rates were investigated. Furthermore, I assessed the exchange rates among alkanethiols with different carbon chain lengths and different size of ethylene glycol units. Interestingly, during performing this research, I found that tetrahydrofuran (THF) highly compromised the monolayer stability, and thus, induced unusually fast exchange of alkanethiolates of SAMs with other alkanethiols in the solution. As such, the use of THF for preparing matrix solution facilitated the desorption of alkanethiolates from the gold and co-crystallization with matrix molecules, and therefore, improved the SAMDI efficiency. I assessed SAMDI efficiency for the analysis of carboxylic acid-, maleimide-, peptide-, biomacromolecule-presenting SAMs using the various organic matrix solutions. In chapter II, a method of small molecule analysis using a converted graphene-like monolayer (CGM) plate and LDI-TOF MS without organic matrices is described. The CGM plate was prepared from SAMs of biphenyl-4-thiol on gold using electron beam irradiation followed by an annealing step, and utilized for the LDI-TOF MS analyses of various small molecules and their mixtures, e.g., amino acids, sugars, fatty acids, oligoethylene glycols, and flavonoids. The CGM plate afforded high signal-to-noise ratios, good limits of detection (1 pmol to 10 fmol), and reusability for up to 30 cycles. As a practical application, the enzymatic activity of the cytochrome P450 2A6 (CYP2A6) enzyme in human liver microsomes was assessed in the 7-hydroxylation of coumarin using the CGM plate without other purification steps. In chapter III, a fast and sensitive method for multiplexed detection of miRNAs is described by combining mass signal amplification and isotope-labeled signal reporter molecules. In this strategy, target miRNAs are captured specifically by immobilized DNAs on gold nanoparticles (AuNPs), which carry a large number of small molecules, called amplification tags (Am-tags), as the reporter for the detection of target miRNAs. For multiplexed detection, I designed and synthesized four Am-tags containing 0, 4, 8, 12 isotopes so that they had same molecular properties but different molecular weights. By observing the mass signals of the Am-tags on AuNPs decorated along with different probe DNAs, four types of miRNAs in a sample could be easily discriminated, and the relative amounts of these miRNAs could be quantified. The practicability of our strategy was further verified by measuring the expression levels of two miRNAs in HUVECs in response to different CuSO4 concentrations.
