The purpose of this study is to verify the physiological activity, potential as a cosmetic material, and industrial applicability of Astragalus membranaceus (A. membranaceus; AM) ethanol and A. membranaceus bioconversion (AMB) extracts. To demonstrate the purpose, the following tests were performed: ① Measurement of anti-oxidative activity, ; ② Assessment of anti-inflammatory effects on RAW264.7 cells, ; ③ Evaluation of melanin biosynthesis inhibition in B16F10 melanoma cells, ; and ④ Determination of the influence of MMP-1 manifestation in HDF cells. Initially, 70% ethanol was used to acquire AM extract, and by the addition of accellerase, AMB extract was acquired. As a result of comparing both extracts’ anti-oxidative activities, the anti-oxidative activity of AM extract was observed to be higher than that of AMB extract. And it confirming the possibility of employing AM exstract as a cosmetic material. To measure the anti-inflammatory influence on RAW264.7 cells, AM and AMB extracts were treated through their concentration. It was then confirmed that the inhibitory impact on NO production was dependent on concentration, and that AMB extract exhibited profound inhibition of NO production, even at low concentrations. As a consequence of measuring ability of melanin biosynthesis inhibition in B16F10 melanoma cells, both of extraction methods were found to possess high melanin bio synthesis inhibitory effects. In addition, the AMB extract was established to be superior to arbutin, the positive control group, in terms of inhibiting melanin biosynthesis, and its inhibitory effect on MITF expression was confirmed. The impact of HDF cells on MMP-1 expression was examined. It was confirmed that each sample had a concentration-dependent inhibitory effect on MMP-1 expression, and it was also verified that the AMB extract inhibited MMP-1 expression through the p-JNK signaling pathway. The results that presented above confirmed the anti-oxidative, and anti-inflammatory effects as well as melanin biosynthesis inhibition of AM and AMB extracts, rendering them as functional cosmetic materials.
