Glutamine is an important amino acid involved in many biochemical pathways. Glutamine metabolism is dysregulated in many cancers which mostly display glutamine addiction for cell proliferation. Here I investigate the function of tumor necrosis factor receptor-associated protein 1 (TRAP1) through its inhibition treatment, gamitrinib-triphenylphosphonium (G-TPP) on glutamine metabolism of non-small cell lung cancer (NSCLC). I found that even harboring the same expression of TRAP1, glutamine-dependent cells showed significant susceptibility with TRAP1 inhibitor treatment compared to the corresponding glutamine-independent group of cell lines, suggesting a potential mechanism by which TRAP1 regulates glutamine metabolism in lung cancer. I then focused on TRAP1-mediated activity of glutamine metabolism enzymes, which revealed that TRAP1 may inhibit glutamine synthetase (GS) enzymatic activity as treatment of G-TPP increased this activity in a dose-dependent manner. Inactivation of GS in NSCLCs may lead to abundance of glutamate entering the TCA cycle which increase ATP generation from glutamine-derived sources and promotes cell growth. Taken together, these findings provide a better understanding of glutamine metabolism in lung cancer mediated by TRAP1 as well as evidence that TRAP1 could be a promising therapeutic target for glutamine addicted cancer cells. My study investigated the regulation of ferroptosis in two subtypes of glioblastoma (GBM), proneural and mesenchymal which are mutually exclusive. By examining the cell viability under amino acids deprivation condition, I found that mesenchymal GBM is dramatically sensitive with cystine deprivation indicating the ROS-mediated cell death. Treatment of ferroptosis inhibitors ferrostatin-1 or deferoxamine thus defined cystine deprivation-induced mesenchymal GBM cell death as ferroptosis form. I further focused on gene expression of ferroptosis regulators, especially lipid and iron metabolism genes to better describe the sensitivity of mesenchymal GBM subtype with ferroptosis. Some ferroptosis inducers are reported to defeat GBM resistance with classic chemotherapy. As therapy resistance is featured characteristic of mesenchymal GBM, I suggested the usage of ferroptosis inducers for personalized therapy in mesenchymal GBM group of patients.
