The bovine viral diarrhea virus (BVDV) belongs to the genus Pestivirus of the Flaviviridae family. BVDV infection causes major economic losses for the livestock industry worldwide. Autophagy is an essential control process by which cells degrade unnecessary or dysfunctional cellular components or organelles in the cytoplasm. In this study, we explored autophagy activity in non-cytopathic (ncp) BVDV2-infected Madin-Darby bovine kidney (MDBK) cells. Using transmission electron microscopy, we showed morphological characteristics of autophagosomes as a result of ncp BVDV2 infection. In addition to this, upregulation of autophagy related proteins, including LC3, ATG5, and ATG14, was also detected by immunofluorescence and western blotting. Taken together, these results suggest that ncp BVDV2 infection activates autophagy in MDBK cells. However, expression of Beclin-1, which is an important factor in the autophagic signaling pathway, was not increased. Though these results will assist researchers aiming to prevent diseases and infection of viral pathogens, further studies are still necessary to clarity the mechanism of autophagy in ncp BVDV2 infection.
