The utilization of renewable feedstock-derived raw materials and development of bioprocesses thereof to substitute petroleum-based products in polymer industry has shown a steep surge in recent years due to concern of future shortage of petroleum resources and in consideration of green production. Compared to chemical catalysis, biocatalysis has countless advantages including high efficiency, increased stability, high degree of selectivity (regio-, chemo-, and enantio-selectivity), and safe reaction conditions. In modern life, biosynthesis allow us continued access to conventional petroleum-based plastics while avoiding harmful environmental effects, and becoming popular due to their biodegradability, sustainability and unique functionality. Exhaustive scientific efforts have established the protocols for large-scale production of low-molecular weight bioplastic monomers (e.g., lactic acid, succinic acid, adipic acid, and putrescine) using green processes. However, it remains a challenging task to produce medium- to long-chain bioplastic polymer monomers (e.g.,ω-hydroxycarboxylic acids, α,ω-dicarboxylic acids, α,ω-diols and ω-aminocarboxylic acids) that exhibit similar or superior physicochemical properties to polyethylene and other bioplastics on an industrial scale by eco-friendly methods. Biochemicals from vegetable oil derivatives (e.g., free fatty acids (FFAs)) are of great interest to the chemical industry as a source for the production of various polymer intermediates and precursors of polyamides and polyesters. It has been reported that omega amino dodecanoic acid (ω-AmDDA) are convenient monomers for Nylon 12 biosynthesis. Furthermore, α,ω-diols are used as monomers for the production of polyesters and polyurethanes. At present, industrial-scale preparation has only been confined to short-chain diol biosynthesis, while biosynthesis of medium- to long-chain α,ω-diols (C8–C16) has not been studied extensively. Terminal hydroxylation, activation and oxidation of C-H bonds of FFAs for the production of ω-hydroxy fatty acids (ω-OHFAs), is however still chemically challenging. It requires rough conditions, high pressure, high temperature and the processes still suffer from poor selectivity. The versatile cytochrome (CYP) P450, enzymes are capable of catalyzing intricate reactions such as regio- and stereoselective oxidation of non-activated FFA’s C─H bonds to the corresponding hydroxy (C─OH) products. Recently, the bacterial CYP153A subfamily of P450, including CYP153A16 from Mycobacterium marinum, CYP153A33 from Marinobacter aquaeolei, CYP153A34 from Polaromonas sp., CYP153A13 from Alcanivorax borkumensis SK2, and CYP153A35 from Gordonia alkanivorans, have been reported as promising biocatalysts due to their functional expression in the soluble form in E. coli and higher regioselectivity (>95%) for the ω-position. In this study, we functionally characterize a novel CYP153A from Mycobacterium parascrofulaceum (MprCYP153A) for the production of ω-OHFAs. We also develop 10 mutants of novel MprCYP153A based on in silico analysis and side directed mutagenesis to find out fatty acid recognition sites and increase ω-OHFAs productivity. Next, we produce ω-AmDDA from dodecanoic acid (DDA), by using the cascade of novel P450 (MprCYP153A), alcohol dehydrogenase (AlkJ) from Pseodomonus putida GPo1, and ω-transaminase (ω-TA) from Mesorhizobium loti. Additionally, ω-AmDDA is also produced using the cascade of artificial self-sufficient P450s instead of CYP153A. The biosynthesis of α,ω-diols (C8–C16) from renewable FFAs is also reported in this study. For the production of medium- to long-chain α,ω-diols from FFAs, first, hydroxylation of FFAs are carried out using CYP153A33 from Marinobacter aquaeolei (MaqCYP153A33). Next, a catalytically efficient carboxylic acid reductase (CAR) from Mycobacterium marinum (MmCAR) recently reported is used for selective reduction of carboxylic acid functional group of ω-hydroxy FFAs to corresponding aldehydes, and endogenous aldehyde reductases (ALRs) of host E. coli cells are used to convert ω-hydroxy fatty aldehydes to α,ω-diols. In summary, the results of this study demonstrated the scope of the potential cascade reaction of CYP P450, ω-TA, CAR and ALR for the production of industrially important monomers, from FFAs. Furthermore, the findings of this study suggested the scope of the potential development of bioprocesses to substitute the petroleum-based products in the polymer industry.
