전통 발효주의 제조 시 전처리법에 따른 누룩의 당화력의 차이를 확인하기 위하여 고문헌에 사용되어진 전처리법 죽, 범벅, 설기떡, 구멍떡, 물송편, 인절미, 개떡, 고두밥으로 단양주를 제조한 후 효소 처리를 하여 당의 함량을 측정하였다. 전통누룩에 존재하는 당화효소가 당화력에 미치는 효과를 확인하기 위해 α-, β-, glucoamylase의 효소활성을 측정하였고, 8가지 방법으로 전처리한 쌀에 효소 무처리, α-, β-, glucoamylase을 각각 처리한 것과, 세 가지 효소를 병행 처리한 총 5가지 방식으로 효소처리를 하였다. α-amylase 효소에 의해 생성된 maltose의 경우 반응직후에는 개떡과 설기떡의 생성량이 상대적으로 많았고, 고두밥에서의 생성량이 가장 적었으나, 48시간 후에는 인절미에서 134.65mg/mL로 가장 많았고, 죽에서 107.59mg/mL로 가장 적은 것으로 나타나 전처리법에 따라 생성량의 차이를 보였다. β-amylase 처리 구에서의 48시간 후 maltose 생성량은 범벅에서 149.13mg/mL로 가장 많았고 고두밥에서 103.74mg/mL로 가장 적은 생성량을 보였으나 α-amylase, β-amylase의 모든 처리구에서 100mg/mL 이상의 maltose가 생성되어 큰 차이를 보이지는 않았다. Glucoamylase 처리 구에서는 α-amylase, β-amylase 처리구와는 다르게 전처리 방법 중 고두밥에서 9.78mg/mL로 최대 생성량을 보였다. 전통 누룩과 유사하게 α-, β-, glucoamylase를 병행 처리한 경우는 고두밥에서 66.69mg/mL로 glucose 생성량이 가장 많았다. 전처리법에 따른 당화의 차이가 알코올 발효에 미치는 영향을 확인하기 위하여 전처리 방법 별로 단양주를 제조한 후 25℃에서 8일간 발효하면서 알코올 함량을 조사하였다. 설기떡으로 빚은 술이 발효종료 시 8.9%로 알코올 생성 최대치를 나타내었으며, 설기떡> 개떡> 인절미> 물송편> 구멍떡> 고두밥> 죽> 범벅 순으로 전처리법에 따라 알코올 생성량에서도 차이를 보였다. 쌀의 전처리 8가지 방법을 이용하여 각각 밑술과 덧술을 제조한 64가지 조합을 구성하여 담금을 실시하였다. 알코올 함량은 밑술을 인절미로 하고 덧술을 범벅으로 하였을 때 18.97%로 가장 높게 나타났으며, 밑술을 범벅, 인절미, 고두밥으로 전처리한 경우와 덧술을 범벅, 구멍떡, 물송편으로 전처리한 경우에 높은 알코올 함량을 보였다. pH는 담금 직후 설기떡(밑술)-고두밥(덧술)이 5.4로 가장 높았고 죽(밑술)-설기떡(덧술)이 4.4로 가장 낮았다. 발효 종료 후에는 모든 발효주가 4.34~4.75 범위 내에서 큰 차이가 없었다. 총산은 고두밥으로 전처리한 조합이 0.04%로 가장 낮았고 범벅(밑술)-물송편(덧술) 조합에서 0.321%로 가장 높았다. 가용성 고형분 함량은 발효 초기에는 고두밥으로 덧술 한 경우가 가장 낮았고, 발효 종료 후에는 죽으로 덧술 한 경우가 모두 낮게 나타났다. 반면 구멍떡, 물송편, 개떡으로 덧술 한 경우 총산 함량이 상대적으로 높은 경향을 나타내었다. 또한 술의 맛에 영향을 주는 아미노산도, 유리당, 유기산 분석을 실시하였다. 아미노산도는 범벅과 고두밥으로 밑술하고 고두밥으로 덧술한 술에서 0.23%로 가장 낮았으며, 밑술의 전처리법에 상관없이 덧술을 범벅으로 제조하였을 때 아미노산도가 높았으며, 범벅(밑술)- 구멍떡(덧술)으로 제조한 술에서 0.76%로 가장 높았다. 그러나 전반적으로 0.3-0.4% 범위 내에서 비슷한 결과를 보였다. 유리당은 발효 잔당으로 주로 존재하는 glucose와 maltose가 검출되었다. 전체 유리당 함량은 덧술을 물송편, 구멍떡, 개떡으로 만든 술에서 높게 나타났다. 유기산의 경우는 succinic acid, lactic acid, malic acid가 전체 유기산 함량의 약 80% 이상을 차지하였으며, 그 중 감칠맛이 특징인 succinic acid의 함량이 가장 높았다. 64종 발효주의 향기성분으로 alcohol 12종, ester22종, acid 8종, aldehyde 1종 등 모두 43종이 검출되었다. 모든 발효주에서 에탄올 외에 과일향을 나타내는 ethyl decanoate와 꽃향, 꿀향을 나타내는 benzene ethanol, creamy하고 milky한 향을 나타내는 ethyl hexadecanoate는 공통적으로 검출된 향기성분이었다. 나머지 성분들은 각 발효주에 따라 상당한 차이를 보였다. 관능평가에서 밑술과 덧술의 전처리법에 따라 유의적인 차이가 나타났다. 밑술을 설기떡으로 제조한 술은 복합미(taste overall)와 종합적인 평가(overall)에서 유의적인 차이가 있었다. 인절미로 밑술을 제조한 술은 신맛에서 유의적인 차이가 있었다. 총기호도(overall)는 냄새(odor)와 맛(sweet, bitter, sour, taste overall)과 후미(mouth feel)와 상관성이 높은 것으로 나타났으며 단맛이 복합미에 큰 영향을 미치는 것으로 나타났다. 쌀의 전처리법을 달리하여 제조한 발효주 64개 조합의 이화학적 특성과 관능적 특성을 전처리법에 따른 밑술과 덧술을 설명변수로 설정하여 분석하였다. 이화학적 특성 중 알코올, 아미노산(%), latic acid은 밑술 보다 덧술에 더 유의적으로 나타났다. 관능적 특성 중 신맛은 밑술에, 시각, 후각, 단맛, 복합미, 종합적인 평가는 덧술에 더 유의적인 것으로 나타났으며, 쓴맛은 밑술과 덧술 모두에 유의적이지 않았다. 주성분 분석(PCA)에서 밑술 보다 덧술이 이화학적 특성에 더 강한 상관관계를 보이는 것으로 나타났고, 밑술과 덧술에 대한 휘발성 향기성분은 이화학적 특성보다 상관관계가 뚜렷하지 않았다. 이화학적 특성과 관능적 특성 간의 상관관계분석에서 알코올은 총산, 당도, 아미노산도, 단맛, 신맛과 종합적인 관능평가 항목에서 강한 상관관계에 있었으며, 관능적 특성 상호간에도 강한 상관관계를 가지는 것으로 나타났다.

To observe saccharification power of traditional Nuruk, rice pre-treatment methods - Juk, Beombeok, Seolgitteok, Gumeongtteok, Mulsongpyeon, Injeolmi, Gaetteok and Godubap – were used to produce single brewed liquor and the amount of reducing sugar was measured. Enzyme activities of traditional Nuruk’s saccharogenic amylase(α-, β- and glucoamylase) were measured. Pre-treated rice was treated in five different methods: control group, α-, β-, glucoamylase and concurrent treatment of the three amylase. 48 hours after mashing, the amount of maltose produced by α-amylase was the greatest at 134.65mg/mL when Injeolmi was used, whereas Juk produced the least amount of maltose 107.59mg/mL. 48 hours after β-amylase was added, Beombeok produced the most amount of maltose 149.13mg/mL, whereas Godubap produced the least amount 103.74mg/mL. Overall, α-amylase and β-amylase treatment both produced more than 100mg/mL of maltose. Unlike α-amylase and β-amylase treatment, glucoamylase created the most amount of maltose, 9.78mg/mL, when Godubap was used. When α-, β-, and glucoamylase were treated concurrently, Godubap produced the most amount of glucose 66.69mg/mL. To observe effects saccharification level caused by different rice pre-treatment on alcohol contents, single brewed liquors were fermented for 8 days at 25℃ and the alcohol contents were measured. Liquors made with Seolgitteok had the highest alcohol contents of 8.9%; the order of alcohol contents from the greatest to the least was Seolgitteok > Gaetteok > Injeolmi > Gumeongtteok > Mulsongpyeon > Godubap > Juk > Beombeok. The alcohol contents were different depending on the type of pre-treatment methods used. 64 combinations of 8 different pre-treatment of mitsul and dutsul were used to produce double brewed liquors. The alcohol contents of liquor was the highest at 18.97% when Injeolmi(mitsul) and Beombeok(dutsul) were used. In general, Beombeok, Injeolmi and Godubap(mitsul) and Beombeok, Gumeongtteok, Mulsongpyeon(dutsul) created liquor with high alcohol contents. The pH level immediately after mashing was the highest at 5.4 when Seolgitteok(mitsul) - Godubap(dutsul) combination was used whereas Juk(mitsul) - Seolgitteok(dutsul) resulted in the lowest level at 4.4. Post-fermentation pH level of all the liquors were between 4.35 and 4.75. Total acid was the lowest at 0.04% when Godubap pre-treatment was used and it was the highest at 0.321% when Beombeok(mitsul) - Gumeongtteok (dutsul) combination was used. Total acid had a tendency to increase as fermentation progressed, but did not show any abrupt changes. Soluble solide contents level in the early phase of fermentation was low when Godubap(dutsul) was used, and post-fermentation soluble solide contents was low when Juk(dutsul) was used. In general, soluble solide contents level was high when Gumeongtteok, Mulsongpyeon, Gaetteok(dutsul) were used. In addition, analysis on amino acid, free sugar, organic acid that influence liquor taste was conducted. Amino acid(%) was the lowest at 0.23% when Beombeok, Godubap(mitsul) - Godubap(dutsul) combination was used. Amino acid(%) was generally high when dutsul with Beombeok was used, regardless of the type of mitsul used. Beombeok(mitsul)-Gumeongtteok(dutsul) resulted in the highest level of Amino acid at 0.76%, but the content level remained between 0.3-0.4%. Glucose and matlose, which are usually products of residual sugar, were detected from free sugar. Free sugar contents were high when dutsul with Gumeongtteok, Mulsongpyeon or Gaetteok was used. Succinic acid, lactic acid and malic acid consisted of more than 80% of organic acid content, among which succinic acid - characterized by its umami - content was the highest. From the 64 combinations of liquor, total of 43 types of volatile flavor compounds were detected; 12 from alcohol, 22 from ester, 8 from acid and 1 from aldehyde. In all liquors made, ethanol content was the highest. Ethyl decanoate(fruity aroma), benzene ethanol(flower and honey aroma) and ethyl hexadecanoate(creamy and milky aroma) were common elements detected in all the liquors. Other flavor compounds had significantly different contents level depending on the pre-treatments used. Sensory evaluation of liquors with 64 different rice pre-treatment variations showed statistically significant difference (p<0.05). Traditional liquor made with Seolgitteok(mitsul) showed a significant difference between overall and overall taste preference. Liquors produced with Injeolmi(mitsul) had a significant difference in sour taste. Although it was not statistically meaningful, this study has confirmed that sensory characteristics of liquors were not significantly influenced by pre-treatment method and that overall preference of a liquor is influenced more by liquor’s taste (sweet, bitter, sour) and mouth feel than its appearance and odor. Physiochemical and sensory properties of 64 combinations of rice pre-treatment were analyzed with mitsul and dutsul as explanatory variables. Among the physiochemical properties, alcohol contents, amino acid(%) and latic acid, variable dutsul is more influenential. Among the sensory properties, sourness had a more significant correlation to mitsul; appearance, odor, sweetness, taste overall and overall had a more significant correlation to dutsul; bitterness had no significant correlation to mitsul and dutsul. Principal component analysis(PCA) showed that dutsul had greater influence to physiochemical properties than mitsul; both dutsul and mitsul had less significant influence on volatile flavor compounds than on physiochemical properties. Correlation analysis on physiochemical and sensory properties showed that alcohol had a significant correlation to total acid, brix, amino acid(%), sweetness, sourness and overall; sensory properties had strong mutual correlations.
