Fc 융합이 중동호흡기증후군 코로나바이러스 항원의 면역원성에 영향을 미치는지 확인하기 위해 우리는 배큘로바이러스 발현시스템을 이용하여 인간 면역글로불린 G4 Fc 영역이 글라이신/프롤린 링커를 통해 중동호흡기증후군 코로나바이러스 스파이크 단백질 (S770)에 결합된 Fc 융합 중동호흡기증후군 코로나바이러스 스파이크 단백질 (S770-Fc, 110 kDa)을 제작했다. 비교 분석을 위해, 면역글로불린 G4의 Fc 영역이 없는 S770 단백질 (S770-ΔFc)을 단백질 분해 효소를 사용하여 생성하였으며, 두 단백질 (S770-Fc, S770-ΔFc)을 쥐에게 근육 주사하여 면역원성을 확인했다. 예상했던 것 과는 달리, S770-ΔFc가 S770-Fc보다 높은 면역원성을 보였다. 또한, S770-ΔFc와 달리 S770-Fc의 접종은 중동호흡기증후군 코로나바이러스에 대한 중화 항체를 유도하지 못했다. 결과를 종합해 보면, 면역글로불린 G의 Fc 융합이 항체 결합 능력을 억제함으로써 S770의 면역원성을 감소시킨다는 것을 나타낸다.

Many studies have shown that the fusion of IgG F domain in vaccine design increases the immunogenicity of various vaccine antigens. However, only a few studies have shown potential negative effects of the Fc fusion on vaccine antigens. To determine whether the Fc fusion affects the immunogenicity of MERS-CoV antigen by using baculovirus expression system, we constructed an Fc fused MERS-CoV spike protein (S770-Fc, 110 kDa), a human IgG4 Fc domain fused to a MERS-CoV spike protein (S770) via a Gly/Pro linker. For a comparative analysis, S770 proteins lacking the IgG4 Fc domain were generated by using the IdeS protease (S770-ΔFc) and the immunogenicity of the above constructs was examined by following intramuscular immunization in mice. Contrary to expectations, non-Fc spike protein (S770-ΔFc; 90 kDa) showed higher immunogenicity than the Fc fusion protein (S770-Fc). Moreover, non-Fc spike proteins elicited neutralizing antibodies against MERS-CoV, while S770-Fc immunization didn’t. Taken together, our results indicate that IgG Fc fusion reduces the immunogenicity of S770 by inhibiting antibody binding capacity.
