The seven strains of E. coli O157:H7 isolated from diarrheal patient's stool, were serotyped and analyzed for plasmid DNA profile, antibiotic sensitivity, and reactivity to VT1, VT, eae, bfpA, EAF genes by the PCR method and DNA probe hybridization, and analyzed by an RAPD. The following results were obtained. 1. The seven strains belonged to serotypes O157:H7, and all strains were positive in the verocytotoxicity test. 2. For plasmid profiles, 7 strains had 60 MDa plasmid and several smaller sizes of plasmid. The plasmid profile typing of E. coli O157:H7 showed 7 types. Plasmid profile type is valuable for the epidemiological examination as a marker. 3. PCR using specific primers for VT1, VT2, eae, bfpA, and EAF gene was tested, all strains carried the eae gene, and Japanese strain J125 carried VT2 only, a Korean strain K157 carried VT1 gene only. The other strain were produced both VT1, VT2 producers. 4. As for the 7 strains of E. coli O157:H7, 100% was resistant to amikacin and oxacillin, but 100% was sensitive to kanamycin, trimethoprim sulfamethozone and gentamycin. The antibiogram typing showed 2 groups. Antibiogram type 1 was predominant with 71.4%. 5. The seven strains could be differentiated by chromosomal DNA fingerprint 7 patterns. The arbitrarily-primed PCR of the strains were diverse, suggesting that the strains may have originated from diverse sources. The chromosomal DNA fingerprinting is useful for the epidemiological study.
