Metabolites determined were 4 primary metabolites; MEHP, MnBP, MiBP, MEP and 2 secondary metabolites of DEHP; 5-OH-MEHP), 5-oxo-MEHP. We analyzed children's urine from 30 boys and 30 girls. The method detection limit of phthalate metabolites were 0.03 ng/mL for MEP, 1.05 ng/mL for MBP, 0.22 ng/mL for MEHP, 0.15 ng/mL for 5-OHMEHP and 0.16 ng/mL for 5-oxo-MEHP, respectively. Switching Column LC/MS/MS was proven to be a useful tool to determine metabolites of phthalate diesters in human urine. The correlation among phthalate metabolites was very high and statistically significant, except MEP. The children's age (months) was negatively correlated to the concentration of phthalate metabolites. The geometric mean concentration of phthalate metabolites (mg/g creatinine) in children's urine were 25.5 for MEP, 130.3 for MnBP, 56.8 for MiBP, 19.5 for MEHP, 85.6 for 5-OH-MEHP and 83.1 for 5-oxo-MEHP, respectively. Levels of estimated daily intake of parent phthalate compounds (${\mu}g$/kg bw/day) were 0.8 for DEP, 5.0 for DnBP, 1.9 for DiBP and $8.9{\sim}14.2$ for DEHP, respectively. Estimated daily intake for DEP and DiBP were lower than those of other studies but the value for DEHP was higher than that of other study.

제안 방법
가수분해된 시료 중의 프탈레이트 대사체가 쉽게 분리될 수 있도록 10 분간 초음파로 처리한 후, 시료 : 아세토니트릴=1 : 2의 비율로 담아 잘 혼합한 후 단백질과의 분리를 위해 원심분리(5000 g, 10 min)하였다.
가장 먼저 용출되는 MEP를 따로 분리하고, 5-OH-MEHP, 5-oxo-MEHP, MiBP, MnBP는 다음으로 스윗칭하였으며, 마지막으로 배출되는 MEHP를 분리하여 스윗칭하는 방법을 선택하였다(표 1).
지금과 같이 플라스틱 제품의 사용이 필수적이고 도처에서 프탈레이트에 노출될 수 있는 상황에서 노출원이나 경로를 더 많이 가지는 어린이의 프탈레이트 노출실태에 관한 면밀한 조사가 필요하다. 본 연구는 스윗칭컬럼을 이용하여 전처리를 간편하게 한 분석방법을 적용하여 초등학생을 대상으로 소변 중 6종의 프탈레이트 대사산물의 농도를 분석하고 내적 노출량을 조사하였다.
본 연구에서는 스윗칭컬럼 방법을 활용하여 소변 중 6종의 프탈레이트 대사산물을 LC/MS/MS를 이용하여 분석하였다.
분석 대상물질인 프탈레이트 대사체의 경우 극성의 차이가 커서 스윗칭 시간을 구분하여 진행하였다.
스윗칭 컬럼은 동시전처리(on-line cleanup) 목적의 크기배제(size exclusion) 방식의 전처리컬럼(MF C8, 4.6×50 mm, 5 μm, Shiseido)과 농축을 위한 컬럼(Cadenza C18, 2.0×30 mm, 5 μm, Imtakt), 그리고 피크의 분리를 위한 분석컬럼(Cadenza C18, 2.0×75 mm, 3 μm, Imtakt)을 스윗칭 밸브를 이용하여 교차하여 통과할 수 있도록 하여 진행하였다(그림 2).
어린이들의 소변에서의 프탈레이트 대사산물 농도를 이용하여 1일 노출량을 추정하였다.
소변시료는 프탈레이트의 오염이 일어날 수 있는 플라스틱 제품을 배제하기 위하여 뚜껑이 있는 10 ml 유리관을 이용하였다. 연령, 가족관계와 같은 일반적인 특성 및 프탈레이트에 노출될 수 있는 생활환경과 식품 등에 관한 정보를 얻고자 설문조사도 병행하여 실시하였다.
전처리를 위해 냉동보관 된 시료를 실온에서 해동하고 완전히 혼합시킨 후, 1.5 ml glass vial에 시료 0.5 ml, 1 μg/ml 농도의 내부표준물질 10 μl 및 βglucuronidase(E. coli 12, Bachem) 10 μl와 1 M 암모늄아세테이트 용액 100 μl를 첨가하여 37oC로 맞춰진 항온배양기에서 2시간 동안 반응시켜 프탈레이트 대사산물의 결합체를 가수분해하였다.
프탈레이트 대사체 및 13C로 라벨링된 내부표준물질의 질량분석기 조건을 확립하기 위하여 주입기(10 μl/min, Harvard Apparatus)로 최적화 과정을 수행하였다.
프탈레이트 대사체 분석을 위하여 기포제거기, 시료주입기를 초음파로 세척할 수 있는 자동시료주입기와 2쌍의 b inary 펌프, 가열기 및 스윗칭 밸브가 장착된 HPLC(Nanospace SI-2, Shiseido)를 이용하였다.
프탈레이트 대사체의 컬럼을 이용한 분리는 질량분석기를 이용하였기 때문에 분석감도를 최적화하는 것에 주안점을 두었다.
하지만, MiBP와 MnBP의 경우 분자량이 같고 구조가 비슷하기 때문에 질량분석기에서의 겹침현상이 나타나 최대한으로 분리할 수 있도록 크기 배제 방식의 전처리컬럼과 실리카 바탕의 분석컬럼을 적절히 활용하였다.
대상 데이터
HPLC에 사용된 용매인 물과 아세토니트릴은 B&J(Burdick & Jackson, NJ, USA)에서, 완충용액 제조를 위한 시약인 암모늄아세테이트와 포름산은 SigmaAldrich에서 구입하였다.
분석대상물질로 선정된 것은 사용량이 가장 많은 DEHP와 DBP, DEP의 대사산물, 즉 MEP(mono ethyl phthalate), MiBP (mono(iso-butyl)phthalate), MnBP(mono(n-butyl)phthalate), MEHP 등 4종의 프탈레이트 1차 대사산물과 DEHP의 2차 대사산물인 5-oxo-MEHP와 5-OH-MEHP이었다.

원심분리된 시료의 상등액을 아세토니트릴로 세척한 주사기와 실린지필터(nylon, 0.2 μm, Whatman)를 이용하여 여과한 후 분석용 시료로 사용하였다.
위 식⑵에서 MEtotal은 MEHP, 5-OH-MEHP, 5-oxoMEHP 농도의 크레아티닌으로 보정된 몰농도(μmol/g Cr)의 총합을 나타내며 MWDEHP는 DEHP의 분자량, f는 0.442를 이용하였다.
이동상 A의 경우 0.1% 포름산 용액을 이용하였으며, 이동상 B는 0.1% 포름산이 첨가된 아세토니트릴을 이용하였다.
정확도 검증과 추출효율 보정을 위해 13C로 라벨링된 내부표준물질 5종(13C4-MEP, 13C4-MBP, 13C4-MEHP, 13C4-5-oxo-MEHP, 13C4-5-OH-MEHP)을 CIL(Cambridge Isotope Laboratory, MA, USA)에서 구입하여 사용하였다.
프탈레이트 대사체의 분리를 위한 이동상 용매는 섞임성의 문제를 최소화하기 위하여 전처리컬럼과 분석컬럼으로 주입되는 각각의 A 및 B 펌프에서 같은 용액을 사용하였다. 이동상 A의 경우 0.
이론/모형
3 농도수준(저농도, 중농도, 고농도)에서 정확도와 정밀도를 평가하였으며, 구체적인 방법은 미국 산업안전보건연구원(NIOSH)20)과 산업안전보건청(US OSHA)21)에서 권고하고 있는 방법을 사용하였다.
Ion souce는 electrospray ionization(ESI) negative 방식을 이용하였으며 Ion source energy는 -4500 V를 유지하였다.
성능/효과
조사대상인 초등학생의 소변 중 프탈레이트 대사산물의 농도는 MEP를 제외하면 상관계수 0.375~0.975 범위의 상관성을 나타내었고, 연령에 따라 감소하는 경향을 보여주었다.

DEHP 3가지 대사산물의 경우 남자 어린이의 소변 중 농도가 여자 어린이의 소변 중 농도보다 유의하게 높게 나타났다(표 4).
DEHP의 1차 대사산물이 MEHP와 2차 대사산물과의 상관성은 통계적으로 유의하지만 상관계수가 2차 대사산물 간의 상관계수보다는 낮았다(그림 3).
16 ng/ml로 평가되었다. MBP의 검출한계가 1.05 ng/ml가 약간 높기는 하지만 전반적으로 혈액에서 프탈레이트 대사체 검출한계와 비슷한 값을 나타냈으며, 농도증가와 기기반응 간의 상관성에 있어서도 매우 우수한 결과를 나타냈다.
MEHP는 회수율 평균은 97.6%, 통합변이계수는 4.2%였으며, 5-OH-MEHP의 평균 회수율은 102.1%, 통합변이계수는 4.4%였고, 5-oxo-MEHP는 회수율 평균은 99.5%, 통합변이계수 값은 5.9%였다.
MEP는 평균 회수율이 99.1%, 그리고 통합변이계수 값은 9.0%였으나 저농도 수준에서 변이계수 값이 13.9%인 반면 중농도와 고 농도에서는 각각 5.1%, 4.7%로 회수율간의 차이가 다소 크게 나타났다.
따라서 이러한 점을 고려한다면 Takatori의 검출한계와 본 연구에서 산출한 검출한계 값 간에는 별 차이가 없는 것으로 보이며, 이병무가 연구한 소변에서의 MEHP가 본 연구보다 높게 평가된 이유는 두 연구에서 적용한 분석 장비의 기본적인 차이, 즉, 본 연구에서는 LC/MS/MS를 적용하였기 때문에 HPLC-UV 방법에 비해 상대적으로 매우 낮은 검출한계를 보인 것으로 판단된다.
모든 시료에서 6종의 프탈레이트 대사산물이 검출가능한 농도 이상으로 발견되었으며, 어린이와 물질의 종류에 따라 농도의 편차가 크게 나타났다2).
본 연구에 참가한 어린이들은 DEP와 DiBP는 독일 등 외국에 비하여 낮았으며, DEHP는 다른 연구결과에서 나타난 성인 및 어린이의 1일 섭취량 추정치에 비해 다소 높은 것으로 확인되었다.
본 연구에서 분석된 6종의 프탈레이트 대사산물의 농도는 어린이의 연령(개월)이 증가할 수록 낮아지는 경향을 보였다. 이 중에서 MiBP, MEHP, 5-OH-MEHP 및 5-oxo-MEHP의 농도는 연령과 유의한 음의 상관성을 나타내었다.
본 연구에서 사용한 스위칭 칼럼 LC/MS/MS 분석방법의 프탈레이트 대사체에 대한 검출한계는 Kato 등16)이 소변에서의 프탈레이트에 대한 대사체를 LC/MS로 분석하면서 평가한 검출한계 결과(5-oxo-MEHP; 1.2 ng/ml, 5-OH-MEHP; 1.6 ng/ml, MEHP; 1.3 ng/ml)보다 우수한 검출한계를 보였다.
소변에서 평가한 MBP의 회수율 평가결과 3농도수준에서 평균 회수율은 100.0%, 그리고 통합변이계수 값은 4.3%였으며 모든 농도수준에서 변이계수 값이 4.1~4.5%를 나타내었다. MEP는 평균 회수율이 99.

정밀도 평가에서의 통합변이계수는 MEP, MBP, MEHP, 5-OHMEHP, 5-oxo-MEHP가 각각 6.5%, 3.6%, 2.8%, 5.1%, 4.6%로 나타나 반복측정의 일정성이 안정적으로 나타남을 보여주었다.
측정된 소변 중 대사산물의 농도는 17.3~139.8 μg/g Creatinine의 농도 범위를 보였으며 MnBP의 농도가 가장 높았다.
특히 5-OH-MEHP와 5-oxo-MEHP의 상관성은 상관계수 값이 0.955로 상당히 높아서 DEHP의 1차 대사산물인 MEHP에서 동일한 대사과정에 의해 생성된 물질임을 쉽게 알 수 있었다.
