즉, 각각의 시료를 일정량 취하여 15분간 광을 차단하여 암적응시킨 후 100 µmole/m2/sec의 actinic light와 1,500 µmole/m2/sec의 saturation light를 조사하고 Fv/Fm을 비롯한 다양한 형광변수(Fo, Φ'PSII, RFd, qP, NPQ)들을 산출하였다(Table 1). 그리고 형광이미지는 위와 동일한 방법으로 암적응시키고 빛을 조사한 후 방출되는 형광을 CCD 카메라로 포획하였으며(Nedbal et al., 2000), saturation light 조사 후 1,000 ms와 3,960 ms에서 방출되는 형광으로부터 각각 초기형광수율(Fo)과 최대형광수율(Fm) 이미지를 얻었고, 이로부터 광계II 활성(Fv /Fm)의 이미지를 산출하였다.
