Alzheimer’s disease(AD)형 치매의 대표적인 병리학적 원인은 무엇인가? | 치매의 약 60-70%를 차지하는 Alzheimer’s disease(AD)형 치매는 amyloid beta(Aβ)단백질이 뇌에 비정상적으로 침착되어 신경독성을 일으키고, 시냅스 소실, 또는 신경세포 사멸을 유도하여 인지기능 저하와 행동장애로 이어진다고 알려져 있다[3,4]. 이러한 질환의 대표적인 병리학적 원인이라고 알려진 Aβ 단백질은 amyloid precursor protein(APP)라는 단백질이 β-secretase와 γ-secretase에 의해 분해되어 생성되며, 정상인의 경우 Aβ를 생성하지 않는 경로를 통해 비독성의 물질로 대사되지만, 노화 등으로 인해 Aβ의 생성이 증가할 경우 뇌에 침착하여 병변을 초래할 수 있다[5,6]. 오늘날 AD를 치료하기 위한 여러 가지 약물이 개발되었으나 장기간 복용 시 간독성과 같은 심각한 부작용을 초래하기 때문에 보다 안전한 방법을 통해 AD를 예방 및 치료하고자 하는 시도가 활발히 이루어지고 있다[7]. |
