In a hemolysing solution used in the determination of glucose in a whole blood sample, consisting essentially of a lysing reagent and a glycolysis inhibitor which inhibits glycolysis in the hemolysate obtained by contacting the blood sample with the hemolysing solution, the improvement wherein the hemolysing solution consists essentially of (a) a buffer providing a pH of about 6-8; (b) 0.8-3.0 moles/liter of sodium chloride; (c) 1-4 g/l of a surface active agent; (d) 0.5-2 g/l of a chelating agent which in the presence of component (c) serves as a stabilizer against glycolysis in the hemolysate; and, optionally, (e) an effective amount of an antimicrobial agent.
